Abstract

Hypocrellin A (HA), the natural derivate extracted from a special herb in the southern part of China, can act as photosensitive candidate, owing to its high quantum yields of active oxygen species. On the basis of the studies of 1, 4-dihydroxyan thraquinone, hypocrellin A was successfully encapsulated into the titanic nanoparticles (directly encapsulated system), and the molecules absorbed on the outside surface of the porous titanic nanoparticles were transferred into their internal areas (ordered self-assembly system). Comparative studies implied that both two entrapping systems extended their absorption spectrum into longer wavelength and possessed not only promoted water solubility, stability, light stability but also enhanced fluorescence quantum yield, singlet oxygen generation efficiency and photocleavage capacility to calf thymus DNA than those of free HA solution. Especially, hypocrellin A can not be released from titanic nanoparticles, which is beneficial to minimizing phototoxic side effects. In conclusion, all these properities of both two delivery systems make them host tremendous promising feasibility and potentiality to be used in clinical photodynamic therapy.
