Lipid is one of the basic material and life-sustaining activities, as well as one important industrial. At present, animal and vegetable oil can not meet with the demands of people nowadays by the development of population. Microbial oil is a new kind of resources after animal and vegetable oil. A lot of microbial oil is rich in polyunsaturated fatty acids which can lower the serum cholesterol. So they play an important role in the health of human being, which are also raw materials of the biodiesel production. The exploitation of microbial oil can ease the situation of shortage of animal and vegetable oil resource. Therefore, the development and utilization of microbial oils have the practical significance for the edible oil and biodiesel. Microbial oils provide a new way of the oil resources.The paper is based on these points, researched the extraction methods, and identified acid-heating extraction as the extraction methods. Compared the oil productive ability of sixteen yeasts, and determined the Lipomyces tetrasporus Y11562 as the best oil production strain. To improve the production of oil, Lipomyces tetrasporus Y11562 was mutated by UV treatment. Mutant UV5 was obtained after passing primary-screen and multiple-screen. The biomass 21.77±0.33g/L, the oil content 8.07±0.20g/L and the oil productivity 37.05±0.35% was obtained after 96hours flask culturing of UV5.these values were obvious higher than the original strain. After passing on many times, these were no distinct changes on the yield. Fermentation tests show that UV5 has a capability to be improved to industrial strain.We studied fermentation condition in the flask culture of UV5 by one-factor experimental design. when shaking flask fermentation conditions were glucose as carbon source; (NH4)2SO4 as nitrogen source, ratio of carbon to nitrogen at 308; initial pH of 6.5; inoculation proportion of 10%; cultivated temperature at 28℃; cultivated time of 96h,the lipid productivity reached 53±0.23%, the lipid content reached 9.09±0.14g/L.We optimized the culture medium in the flask culture of the UV5 by response surface methodology. The optimal shaking flask culture medium were: glucose at 129.36g/L, (NH4)2SO4 at 0.77g/L, yeast powder at 2.07g/L,KH2PO4 at 1.0g/L,MgSO4·7H2O at 0.5g/L. The lipid productivity was 58.42% theoretically, 57.90±0.50% practically.
