Objective:In order to provide the reference for treatment clinical resistance on e.coli from clinical Fowl and the development of veterinary drugs, the drug resistance inhibitors were screened which possess obviously bacteriostasis through investigating the effect of single extractive and compound extractive of seventeen Chinese medicine drugs on multidrug resistant e.coli.Methods:The multidrug resistant e.coli was screened out to study the bacteriostasis experiment in vitro, by means of separation e.coli, identification serotype and determination resistance. The studies of Chinese medicine drugs extractive on the multidrug resistant e.coli and ATCC25922 were conducted respectively by means of test tube doubling dilution, flat plate punch method and plate punch dilution method. And then the unite sensitization trails of Chinese drugs pharmaceutics which had more bacteriostasis and antibiotic which had less bacteriostasis were conducted in vivo and in vitro respectively. In vitro test, the sensitization effect was observed on resistance e.coli in vivo test, the clinical changes of every fowl were recorded. The dead and morbid chickens were dissected to determin biochemical indicator of serum and observe immune organ indexes.150 60-day-old Saibei Silkies were randomly arranged into 5 groups, the negative control were injected with 0.3ml normal saline by intraperitoneal, the positive control and experiment group I, group II, group III were inoculated with 0.3ml suspension by an intraperitoneal injection, and drenched respectively with the extractive of CHM I 1.5ml、1ml、0.5ml(2g/ml)each day for seven consecutive days. At the day 2,8 after inoculation,4 Silkies were randomly taken from each group for detecting blood serum biochemical indicator and immune organ index.Results:1.15 strains isolated were obtained from the path genetic fowl in five district and identified as 3 serotypes(078, O1和O1+O48) by means of bacteria culture and biochemistry identify. The main serotypes were O1+O148 in this district. The drug resistant test of 20 antibacterial showed that 3 strains of e.coli belonged to multiple resistant.2. The bacteriostasis experiments in vitro indicated that 7 of 17 chinese medicine drugs had obviouslty effect on e.coli, it included goldthread, ash bark, magnoliae cortex, kuhseng, dyers woad leaf, andrographitis, scutellaria. CHM I had very significantly sensitization on resistant e.coli and no effect on ATCC25922. the dose of CHM I was 2g to every 60-day-old Saibei Silkies in vitro one time a day, the inoculated dose was 0.3ml (2.4x1O8CFU) on fowl infected no.l. CHM I had obviously drug reverse effect on resistant e.coli to gentamicin in vivo.3. The experiments indicated that CHM I had very obviously effect on biochemical indicator on Saibei silkies artificial infected resistant e.coli. The results were as follow:①CHM I could promote growth of the immune organs (thymus and bursa of Fabricius) and immunity in silikies.②CHM I could reduce obviously the contents of ALT、AST and GLU in blood-serum, and protect liver and choleretic effect.③CHM I could increase obviously the contents of ALP and Ca2+, and keep calcium balance in blood serum.④CHM I could relieve the harmfulness on kidney, promote food intake and increase weight and regulate the microcirculation on silkies. Conclusions:CHM I had obviously reverse effect on against multidrug resistant e.coli isolated from fowl and immune protection. It had a high popularize value.
