Non-heading Chinese cabbage(Brassica campestris ssp. chinensis Makino) is originated from China, which is one of the important vegetables in eastern Asia and play an important role in daily menu. QTL analysis Vc trait was also carried out. The main results are as follows:1. The suitable concentration of four factors, Mg2+, dNTP, Taq polymerase, and primer were identified by single factor tests in the SRAP amplification system on non-heading Chinese cabbage (Brassica campestris ssp. chinensis Makino), and orthogonal scheme in four factors at three levels was designed based on the result of above single factor test to optimize SRAP amplification system. An ideally SRAP reaction system was established that 25μL reaction volume containing Mg2+ 2 mmol/L, dNTP 0.2 mmol/L, Taq polymerase 1.5U, primer 0.36μmol/L, 1×PCR buffer, and template DNA 30 ng.2. A molecular genetic map of non-heading Chinese cabbage was constructed with a population of 181 F2 individuals from the cross of’Changzhouwutacai’x ’Erqing’. Four kinds of markers of ISSR、RAPD、SSR and SRAP were adopted and 138 polymorphic markers including 4 ISSR,19 RAPD,22 SSR,93 SRAP markers, which was consisted of 11 linkage group and cover 945 cM. A total of 36 (26.1%) distorted markers distributed in the map. The map consists of linkage group, number 138 of markers included in the linkage groups varied from 4 to 33, the length of linkage groups from 53 to 164 cM, and a mean marker interval distance of 6.8cM between markers.3. With composite interval mapping (CIM) method, a molecular genetic linkage map of non-heading Chinese cabbage were adopted to map and analyses QTL controlling the vitamin C trait. Two QTL were detected for controlling vitamin C, Its explained 4.74% and 7.63% of phenotypic variation, respectively. This QTL in non-heading Chinese cabbage were first report, This result is fundamentals for molecular assisted selection of quality-related traits in non-heading Chinese cabbage breeding.
