Abstract

From tissue culture, artificial seed technology, culture and other aspects of physiology and biochemistry Breit for a more systematic study achieved the following main results: In the right Pinellia physiological and biochemical studies to tuber as experimental material, the use of positive post test design Greenhouse and field experiments. Pot experiment mainly studied the light intensity, fertilizer levels, growing media for plant growth and other physiological and biochemical effects. The results showed that: light intensity, fertilizer levels and growing media on the growth and physiological and biochemical Pinellia played an important influence, pot experiments under different conditions, willow-type Breit, triangular blade type Pinellia ternata and peach leaf emergence time to reach 1% germination rate differences were significant, petiole length, petiole diameter, leaf length and leaf width difference reaches 5% significance level, pot experiments under different conditions and peach leaf willow-type type Pinellia tuber yield differences to achieve a % significance level, triangular blade tuber yield differences reached the 5% significance level, willow-type Breit, triangular blade type Pinellia ternata and peach are A 2 , B 1 , C 3 highest tuber yield under the conditions, light intensity, fertilizer levels and growing media three factors on tuber yield has reached a significant level of light intensity on which tuber yield the greatest impact. Pinellia pot experiment different conditions on physiological and biochemical effects are also different: Breit chlorophyll content difference of 5% significance level; Breit soluble sugar content reaches 1% significance level; nitrate reductase is an inducible enzyme, which reflects the use of plant This study found that the ability of nitrogen, pot experiments under different conditions, nitrate reductase activity was significantly difference at 1% level; pot experiments under different conditions, differences in tuber MDA content up to 1% significance level; pot experiments under different conditions Pinellia root activity the difference reached a significant level to 1%. Field experiments, focusing explored mu seed quantity, acres of manure fertilizer, maize intercropping density and frequency of weeding on growth and physiological and biochemical Breit effects. The results showed that: mu seed quantity, acres of manure fertilizer, maize intercropping density, weed growth times on the Breit plays an important physiological and biochemical effects of different field cultivation conditions, Pinellia petiole length, petiole thick degree difference reached a significant level of 1% to the different field cultivation conditions tuber yield differences at 1% level of significance, willow-type Pinellia A 3 B 2 C 2 D 1 highest yield, peach leaf type A 3 B 2 C 2 D 2 Breit highest yield of two leaf types are mu seed quantity Pinellia greatest impact on yield. Breit field cultivation conditions on different physiological and biochemical effects are different: Breit difference chlorophyll content up to 1% significance level; root activity is an objective reflection of root physiological activities of life, different field cultivation conditions Pinellia root vigor differences reach 1% significance level, and peach leaf willow-type Pinellia ternata are type A 3 B 1 C 1 D 3 root vigor maximum treatment conditions, but the number of willow-type Breit weeding greatest impact on root vigor, peach leaf type Pinellia acres manure application rate on root vigor greatest impact. The efficacy of different harvest Pinellia different ingredients, in mid-October guanosine content, total alkaloid content and β-sitosterol content higher than identified as more appropriate harvest period. Reproductive biotechnologies in Pinellia experiment, orthogonal design study hormone concentrations, light intensity, temperature factors on callus induction, bud differentiation, induction of small tubers, roots and other effects. Meanwhile, the preliminary exploration of the Breit artificial seed production technology. The results showed that: in a sterile system to establish the process, when the mercuric chloride concentration was 0.15%, the processing time was 4 min, the Breit blade pollution and mortality rates were 8.8889% and 15.0000%, respectively petiole pollution and mortality of 11.6162% and 9.0909%, better; in callus induction, bud differentiation and small tuber induction process, we found the blade inoculated MS BA 1.0 mg · L -1 NAA 0.5 mg · L -1 medium, the light intensity of about 1000 Lx, under the conditions of temperature 25 ℃, while the petiole inoculated on MS BA 1.0 mg · L -1 NAA 0.2 mg · L -1 medium, the light intensity of about 1000 Lx, temperature 25 ℃ conditions, callus induction rate of up to 100% and the formation of callus better quality; inoculated on MS BA 2.0 mg · L -1 2,4-D 0.5mg · L -1 medium, the temperature 25 ℃, the light intensity of about 2000 Lx, irradiation time 12 h · d < sup> -1 conditions can significantly induce callus buds. The leaves were inoculated in MS BA 0.5 mg · L -1 2,4-D 3.0 mg · L -1 Carbon 0.05% medium, temperature 25 ℃, light strength of about 1000 Lx, irradiation time 12 h · d -1 conditions can significantly induce the occurrence of small tubers. In the Breit rooting in the IBA and BA concentration and ratio were set up and found that the buds were inoculated to MS IBA 1.0 mg · L -1 BA0.1 mg · L - 1 medium, 15 d after the number reached 6.2 Pinellia root, root thick and long, works best. Groping Pinellia transplanting cultivation conditions in tissue culture as experimental materials to study the culture medium and light intensity on the survival rate of transplanted tissue culture, the results showed that: the plantlets were transplanted to the Land: humic soil: perlite = 2:2:1 matrix, placed in the shade under the conditions of 45%, the highest survival rate of 96%. Breit artificial seed production techniques in the study, found in the different treatment under the Breit matrix embedding artificial germination rate difference was significant, of which 4% sodium alginate 0.5% activated carbon as the embedded substrate at a temperature of 25 ℃, Light intensity 1000 Lx conditions germination rate up to 98.5%.
