Abstract

In this study, immunofluorescence staining observed symbiotic system of two arbuscular mycorrhizal fungi acuminata seedlings microtubule structure, to explore the role of microtubules in the establishment of the symbiotic system process; through hi to arbuscular mycorrhizal symbiosis system The tree seedlings applied exogenous camptothecin solution, explore arbuscular mycorrhizal camptothecin and microtubule structure three main results are as follows: 1, observed acuminata seedlings inoculated with arbuscular mycorrhizal fungi in the apical meristematic zone, elongation zone and maturation zone microtubule morphology results acuminata seedlings microtubule-intensive cells in the apical meristem region around the nucleus, dense and strong fluorescence polar microtubule elongation zone cells, and mature District cortical microtubules regularly the oblique parallel structure or spiral mesh structure. Overall acuminata seedlings to mature area apical meristem area, along with cell differentiation and maturation of microtubules in the cell by the initial transverse or oblique ring structure is gradually transformed into a mature spiral mesh structure. Basically consistent with changes in the arrangement of the microtubule people observed herb root tip cells. 2, two arbuscular mycorrhizal that honey-colored sessile capsule mildew (Acaulospora mellea Spain Schenck Am) and root Glomus (Gloums.intraradices Schenck Smith, Gi) single vaccination acuminata seedlings able acuminata seedlings very the good infection symbiotic form arbuscular mycorrhizae. Immunofluorescence staining observed the two mycorrhizal symbiotic system with the infection process of plant cell microtubules structure changes from the initial distribution, microtubule periphery of the spiral structure in the cytoplasm becomes gathered manner aggregates arbuscular around and arbuscular nucleus and cell membrane connected together. Two arbuscular mycorrhizal fungi acuminata seedlings root cortical cells microtubules. By digestion of the fungal cell wall, observed the the two fungi microtubule structure, results show that the External Hyphae within the hyphae and arbuscular bundle microtubules and microtubule from exogenous hyphae endogenous hyphae, arbuscular trunk to fine arbuscular has been continuous, indicating that microtubules may play an important role in the arbuscular mycorrhizal fungal growth. Vesicles was not observed in the structure of the microtubules, but the parts connected by the blister within hyphae bright fluorescence. 3, outside source camptothecin treatment acuminata seedlings inoculated arbuscular mycorrhizal fungi Gi, results showed that the colonization rate and degree of mycorrhizal colonization increased significantly, and significantly increased endogenous camptothecin content. Increase in the content of endogenous camptothecin may be applied is closely related to exogenous camptothecin may be exogenous and camptothecin degree of arbuscular mycorrhizal colonization indirectly increase endogenous camptothecin content. Camptothecin treatment of exogenous acuminata seedlings root cortical cells microtubule arrangement changes, arranged in a helical arrangement becomes random direction. The mycorrhizal symbiotic system of exogenous camptothecin processing plant and fungal microtubules fluorescence intensity is significantly enhanced, and may indicate that the synthesis of a new micro tube.
