Optical coherence tomography (OCT) is a noninvasive morphological method. It allows high-resolution imaging in vivo. An OCT can be used to measure the optical parameters and the structure of tissue. The penetration depth of OCT is sufficient to provide high-resolution images of the epidermis and papillary dermis. As a result, the OCT technology is an effective method to detect the skin noninvasively.Firstly, an introduction to the diagnostic method of skin was briefly reviewed. The application and development of diagnosis based on OCT were reviewed, too. Further more, the purpose of our study was also presented. Based on the skin structure, the mechanism of photoaging and photorejuvenation were introduced. Then combining the instrument we used, the method of observation and estimation was discussed.Secondly, the process of routine histology was observed by OCT. The distance between the entrance peak and the valley and the distance between the entrance peak and the second intensity peak were measured and compared. The effect of physicochemical factors on the thickness of skin was studied.Thirdly, the model of photoaging was provided. The photoaging of mouse skin was monitored by OCT image, biopsy and DIC, and so that the trend of the morphology and optical parameters were obtained.Finally, the mouse skin was irradiated by an IPL system in vivo under certain wavelengths with different energy densities. The injury threshold was determined. Meanwhile, the skin injury recovery process was observed by OCT. The reasons concerning some complications were analyzed by the optical parameters’ changes.The measurement methods and some experimental results are useful for the skin science and the medical purpose.
