Abstract


In present study,we used the benzotriazole compound 2.4-Dichloro-benzoic acid 1-benzotriazol-2-ylmethyl-2-(4-chloro-phenyl)-2-oxo-ethyl ester(Bz) which was one of the zotriazole derivatives.It was demonstrated that Bz could significantly inhibit proliferation of cultured hepatocarcinoma BEL-7402 cancer cells and rat breast 4T-1 cancer cells respectively in a dose-and time-dependent manner,and it could induce the DNA fragmentation.We tested the changes of reactive oxygen species,mitochondrial membrane potential and ATPase in cells. The results suggested that Bz-induced cell growth inhibition and apoptosis may be through modulation of mitochondrial functions regulated by reactive oxygen species.Besides,in 4T-1 cells Bz could cooperate with TZD.Bz of low dose could change the mitochondrial membrane potential and may induce some enzymes activity,which enhanced the efficiency of TZD.We investigated the effect of GSH depletion on mitochondrial function and generation of reactive oxygen species in 4T-1 cells treated with Bz.We used BSO to block GSH synthesis.Mitochondrial membrane potential and the generation of reactive oxygen species were monitored.We provide evidence for a critical role of GSH in determining the susceptibility of 4T-1 cells to Bz through control of mitochondrial oxidative stress.Significantly,Bz may have the anti-cancer potential.
