Objective: To explore sinomenine in vitro induced apoptosis of human cervical cancer Hela cells , in order to found a Inhibition of cervical cancer cell growth by inducing apoptosis of tumor cells to new treatments ; explore Sinomenine apoptosis of cervical cancer Hela the molecular mechanisms of apoptosis , provide laboratory to explore the sinomenine the anti - cervical cancer drug . Methods: logarithmic phase Hela cells after treatment with sinomenine from morphological observation , MTT cell viability determination , flow cytometry analysis detected sinomenine on human cervical cancer Hela cells induced apoptotic effects . the immunoblot (WesternBlot) detection sinomenine treated human cervical cancer Hela cells , COX - 2 , p -Akt , Caspase -3 expression change . In Results: Sinomenine in vitro role of Hela cells , morphological observation the visible cell shrinkage , cytoplasmic condensation , refractive index enhancement , partially adherent cells round shrink to become suspended cells ; MTT assay of Hela vitality of cells , 0.625 mmol / L sinomenine 24 hours after Hela cell growth inhibition , 0.625mmol / L to 2.5mmol / L concentration range , with the increase in drug concentration , Green vine alkali Hela cell growth inhibition rate gradually increases ; flow cytometry analysis of apoptotic cells increased with the increase of the concentration of the drug . Western blot results suggest that the impact the sinomenine of COX-2 and apoptosis -related protein AKT and Caspase-3 expression , with increasing drug concentration , COX-2 and AKT decreased expression of Caspase -3 expression . Conclusion: sinomenine in vitro can significantly inhibit the proliferation of human cervical cancer Hela cells , this inhibition is achieved by Hela cells induced apoptosis , the mechanism may be by inhibiting the expression of COX-2 down Akt protein expression , increased caspase-3 expression work .
