Abstract


Background:As the knowledge of tumorigenesis mechanism being more profound and extensive, scientists have focused on the correlation between epithelial tumors and bone morphogenetic protein 2 (BMP2). It has been proved recently that over expression of Idl (inhibitor of differentiation or inhibitor of DNA binding 1) is related to the growth and malignancy in some types of tumor cells. Studies have demonstrated that over expression of Idl occurs in many malignant tumors. Furthermore, it has been confirmed that BMP2 induces indirectly increased expression of Idl in many tumor cell lines. It is unclear if the expression of BMP2 is correlated with the expression of Idl in prostate cancer. Recently, curcumin has been a focus of the research on prevention and therapy of tumor. It is unclear if curcumin affects the expression of BMP2 and Idl in prostate cancer cells.Objective:To investigate the expression of BMP2 and Idl in primary prostate cancer (PCa) and benign prostatic hyperplasia (BPH) tissues, and its correlation with cellular differentiation. To further analyze the relationship between the expression of BMP2 and Idl in prostate cancer. To observe the effects of curcumin on the gene expression of BMP2 and Idl in androgen-independent prostate cancer PC3 cells, in order to find more effective measures for prostate cancer.Methods:Expression of Idl and BMP2 were respectively detected by immunohisto-chemical streptavidin peroxidase (SP) method in prostatectomy samples of prostate cancer and BPH. The mRNA expression of Id1 and BMP2 were detected by real-time RT-PCR in PC3 cells treated by curcumin at different concentrations.Results:Negative to weak expression of BMP2 and Idl was found in BPH. In contrast, all PCa tissues showed positive expression of BMP2 and Idl. The high-expression of BMP2 and Idl was correlated with Gleason grade in PCa (rs=0.61, 0.63, P<0.01). The expression level of BMP2 was positively correlated with exprssion level of Idl (r=0.92, p<0.01). In PC3 cells, the relative quantity of BMP2 mRNA and Idl mRNA obviously decreased by curcumin. The decreased quantity of BMP2 mRNA and Idl mRNA was correlated with the increased concentration of curcumin (P<0.05). The decreased quantity of BMP2 mRNA was positively correlated with that of Idl mRNA (r=0.85, P<0.01).Conclusion: 1. The expression of BMP2 and Idl in prostate cancer is obviously higher than that in BPH. The high-expression of BMP2 and Idl is positively correlated with Gleason grade in prostate cancer. The expression of BMP2 is positively conrrelated with the expression of Idl in prostate cancer.2. Curcumin can reduce the gene expression of BMP2 and Idl in PC3 cells.
