Expression of RRM1 and MDR1 in Cervical Squamous Cell Carcinoma and Its Relationship with the Chemo-sensitivity to NACT

Abstract


Objective To study the expression of Ribonucleotide Reductase M1 (RRM1) and Multi-drug Resistance 1(MDR1) in cervical squamous cell carcinoma, the relationship of RRM1, MDRl and the relationship with the chemo-sensitivity to NACT.Method Using the immunohistochemistry PV-6000 and RT-PCR, we comparatively studied the expression of RRM1 and MDR1 on 40 cases of cervical squamous cell who accepted NACT. To analyze the expression of RRM1 and MDR1 in cervical squamous cell carcinoma, the relationship of RRM1,MDR1 and the relationship with the chemo-sensitivity to NACT through Pearson chi-square test, Fisher’s exact test, t test and Spearman correlations.Result 1、The result of immunohistochemistry showed:After NACT, the expression of RRMl in drug resistance group was not higher than before (x2=2.763, P=0.096). And the expression of RRM1 in drug sensitive group was no significant change (x2=3.247, P=0.072). The result of RT-PCR showed: after NACT, the expression of RRM1 in drug resistance group was higher than before (t=2.284, P=0.037), but in drug sensitive group was no significant change (t=0.861, P=0.398)2、The expression of RRM1 was correlated with the expression of MDR1 [Before NACT:(the result of immunohistochemistry showed:r=0.802, P=0.000; the result of RT-PCR showed:r=0.377, P=0.016). After NACT:(the result of immunohistochemistry showed:r=0.771, P=0.000; the result of RT-PCR showed:r=0.407, P=0.010)].3、Before NACT, RRM1 negative group of chemotherapy response rate (75.0%) was higher than RRM1 positive effective rate (37.5%) (x2=5.625, P=0.018); the result of RT-PCR showed the expression of RRM1 in drug resistance group was higher than in drug sensitive group (t=2.086.P=0.044)Conclusion RRM1 and MDR1 expressed in cervical cancer together. NACT may raise the level of RRM1 expression, and both of them possibly participated in the process of platinum resistance. So RRM1 should be further investigated as the predictive markers for the chemo-sensitivity to NACT in cervical cancer.
