Objective: To study mdr1 gene methylation levels correlated with cervical cancer chemotherapy; analysis the mdr1 gene methylation levels before and after chemotherapy difference looking for neoadjuvant chemotherapy resistance. Sensitive indicators. Methods: conventional immunohistochemical detection of MDR1 gene encoding P-gp expression levels using MALDI-TOF MS for quantitative analysis of DNA methylation. Results: (1) the expression of P-gp in normal cervical tissue were negative, the effective chemotherapy group before chemotherapy, the expression of P-gp low, high expression of P-gp ineffective chemotherapy group between the two groups was significant difference ( P lt; 0.05). (2) chemotherapy can induce cervical cancer P-gp expression, the expression level was significantly higher than that before chemotherapy group, the two groups have a significant difference (P lt; 0.05). (3) cervical cancer underwent neoadjuvant chemotherapy the sensitive group CpG 2 3,4 sites of methylation is higher than neoadjuvant chemotherapy resistance group. (4) cervical cancer underwent neoadjuvant chemotherapy before the group of CpG 7 , CpG 8 of of CpG 1 2,13, CpG 1 8 , CpG 1 9,20, CpG 2 3, CpG higher than in the line of new 2 4 sites of methylation after neoadjuvant chemotherapy group (P lt; 0.05).
