Objective:The expression of secretend protein acidic and rich in eysteine (SPARC), E-cadherin and the zinc finger transcription factor Snail were detected in nonneoplastic mucosa tissue and gastric carcinoma tissue and discussed the relationship among them. Further research was used to find the relationship between their expression and the invasion, metastasis and prognosis of gastric carcinoma.Methods:①To make 123 cases of gastric carcinoma tissue and 41 cases of nonneoplastic mucosa tissue paraffin specimens in tissue microarray by tissue microarray technology②PV6000 immunohistochemical staining was used to determine protein expression of SPARC, E-cadherin and Snail in nonneoplastic mucosa tissue and gastric carcinoma tissue.③It was using SPSS 16.0 software to analyze those immunohistochemical results of SPARC, E-cadherin and Snail and the relationship between their expression and clinicopathological parameters of patients and survival. P<0.05 for the difference was significance.Results:①In the 123 cases of gastric carcinoma tissue, compared with the results of stromal cells in nonneoplastic mucosa and tumor cells, there were significant difference of high expression rates of SPARC in stromal cells around tumor cells (P<0.05). The high expression of E-cadherin and Snail were significant difference between gastric carcinoma tissue and nonneoplastic mucosa tissue (P all <0.05).②The high expression of SPARC in gastric carcinoma stromal cells was closely correlated with depth of invasion,lymph node metastasis and TNM staging (P all <0.05).The low expression of E-cadherin and the high expression of Snail in gastric carcinoma tissue were closely correlated with depth of invasion, lymph node metastasis and TNM staging (P all <0.05).③The median survival time for patients with high expression of SPARC, E- cadherin and Snail protein expression was statistically significant (p<0.05) compared to patients with low expression of SPARC, E-cadherin and Snail. E-cadherin, depth of invasion, lymph node metastasis and pathological stages were independent prognostic factors by Cox model analysis.④The expression of SPARC and the expression of Snail in gastric carcinoma tissue was positively correlated (P<0.05); the expression of E-cadherin was negatively correlated with the expression of SPARC and Snail (P<0.05).Conclusion①The high expression of SPARC and Snail and the low expression of E-cadherin might be important biological markers for invasion and matastasis of gastric carcinoma. The combined detection of SPARC and Snail and E-cadherin has positive effects on predicting the invasion and metastasis in gastric carcinoma.②The expression of SPARC and Snail and E-cadherin in gastric carcinoma tissue was correlated with patient’ s survival time. Therefore, SPARC, Snail and E- cadherin can be used as indicators for gastric carcinoma prognosis.
