Protein is a very important bioactive substance in organism, it participates in every reaction and activity of organism. The quantitative determination of protein is the foundation of studying protein. So, establishing fast, simple, sensitive and selective methods for determination of protein are very valuable. Studying bioactive substances by electroanalytical chemistry method produces a new field that is bioelectroanalytical chemistry.This thesis is focused on determination of serum albumin and hemoglobin by single sweep polarography. The methods for determination of serum albumin and hemoglobin are established in the presence of dissolved oxygen. The mechanism is studied. Moreover, we study the method of simultaneous determination of hemoglobin and serum albumin by single sweep polarography.The main work and fundamental conclusions are outlined as follows:1. Parallel catalytic wave of serum albumin in the presence of dissolved oxygen A parallel catalytic wave at about -1.85V (vs. SCE) in a supporting solution of 0.08 mol/L pHosphate buffer solution (pH=6.6) was found by using single sweep polarography. Based on this wave, a simple, rapid and reliable polarographic method for serum albumin was developed. The peak height is linearly related to the serum albumin concentration in the range of 0.08~ 4 mg/L. The detection limit for serum albumin is estimated for 0.05 mg/L. The proposed method is applied to determinate serum albumin in sample with satisfactory results.2. Parallel catalytic wave of hemoglobin in the presence of dissolved oxygen and its applicationsA parallel catalytic wave at about -0.62V (vs. SCE) in 5×10-3 mol/LNaOH solution was found by using single sweep polarography. Based on this wave, a simple, rapid and reliable polarographic method for hemoglobin determination was developed. The peak height is linearly related to the hemoglobin concentration in the range of 0.05~ 21 mg/L (correlation coefficient 0.999). The detection limit for hemoglobin is estimated for 0.05 mg/L. The proposed method is applied to determinate human hemoglobin in blood sample and urine sample with satisfactory results. Results indicate that the parallel catalytic wave resulted from the electrochemical reduction of HbFe (III) ion and chemical oxidation of the reduction product, HbFe (II), by dissolved oxygen.3. Simultaneous determination of bovine hemoglobin and bovine serum albumin by single sweep polarographyA method for the determination of bovine hemoglobin and bovine serum albumin simultaneously in a supporting solution of 0.08 mol/L phosphate buffer solution (pH=6.8) in the presence of 1.5% Na2SO3 by single sweep polarography has been established. The results demonstrate that the peak height is linearly proportional to the concentration of bovine hemoglobin in the range of 6~24 mg/L (r=0.996) and 2~8 mg/L (r=0.996) bovine serum albumin and the detection limits are 0.3mg/L for bovine hemoglobin, 0. 1 mg/L for bovine serum albumin when the proportion of bovine hemoglobin and bovine serum albumin are range from 2.5:1 to 5:1. At the same conditions, the concentration of human hemoglobin in the range of 6~33 mg/L (r=0.995) and 2~11 mg/L (r=0.998) human serum albumin can be determined simultaneously and the detection limits are 0.3 mg/L for human hemoglobin, 0.2mg/L for human serum albumin.
