Abstract

Liposomes as a carrier or bioreactor material encapsulation efficiency , good biocompatibility , low toxicity and biological advantage . However , due to the poor stability of the liposomes , industrialization, there are difficulties and other reasons , has listed varieties at home and abroad is still small . Therefore become an important means to improve its performance in the application of liposomes modified liposomes . A study contents of this thesis is the preparation, characterization of modified silica wrapped organic / inorganic composite liposomes . Before modification with the silica inclusions , liposome particle size of about 103 nm , the surface potential of -0.3 mV , silica modified resulting organic / inorganic composite liposome particle diameter of about 127 nm , surface significantly negatively charged, potential about -11.6 mV. TEM observation showed that the deposition of silica in the surface of liposomes formation of nanoscale inorganic silica shell thickness . Another is the preparation of organic / inorganic composite liposomes for the embedding of the anticancer drug doxorubicin . Surface significantly negative electrical potential of about -9.2 mV , transmission electron microscopy shows the formation of nanoscale silica deposition in doxorubicin liposome surface modified silica wrapped organic / inorganic composite liposome particle size of 135 nm or so , inorganic silica shell thickness . Doxorubicin encapsulation efficiency reached 72.4% . Drug release in vitro experiments show good slow release composite liposomal doxorubicin . The study results showed that the silica is used as the protective layer due to the formation of nanometer thickness of the inorganic surface of the liposomes , maintaining the essential characteristics of the liposomes , the stability than conventional liposomes significantly improved and can be used doxorubicin Known as sustained release drug delivery . The surface coverage of the silica has good biocompatibility . Since the silica has a good chemical activity , various targeting molecules can be easily connected to the surface of the composite liposomes , so that the effective delivery and release of the drug to the tumor tissue .
