In 2009-2010, through field production, respectively in the tobacco institute of science of Luzhou and Yibin about different genotypes and different nitrogen to Studying on leaf histological and cellular features of flue-cured tobacco upper leaves during growth and development. Combining indoor slice and electron microscopic analysis .The result showed that:1. Study on histological characteristics of different genotypes of flue-cured tobacco upper leaves during growth and developmentWith the development of leaves,Both Yunyan87 and Zhongyan103, no significant changes in the thickness of the upper and lower epidermis, Thickness of palisade tissue, spongy tissue thickness and leaf thickness increased with the increase of leaf length, Palisade cell density was "∧" type of change,And reached its peak in leaf length 20cm;palisade tissue thickness/sponge tissue thickness、palisade tissue thickness /leaf thickness in the whole growth period with leaf length increased overall upward trend.2. Change of ultra micro structure of different genotypes of flue-cured tobacco upper leaves during growth and developmentWith the development of tobacco ,Palisade cell process significant difference between Yunyan87 and Zhongyan103.Before leaf length 20cm, Palisade cell growth mainly to split, Chloroplasts gradually development,. After 40cm leaf length, Chloroplast structure development is perfect, The cells into the function mainly by synthesis periods. Leaf length 60cm, the cells into accumulation phase with hydrates. When Leaf length setting, hyracoid slice layer reduced, Photosynthetic capacity decreased. Mature leaves, chloroplast degradation, the accumulation of large starch grains and osmiophilic granules.3. Study on histological characteristics of different nitrogen of flue-cured tobacco upper leaves during growth and developmentWith the development of leaves of different treatment, Palisade cell density was "∧" type of change,No nitrogen treatment under the leaf palisade cell development slow, Cell division and elongation growth was inhibited, Intercellular space is also smaller expansion ; Spongy tissue of the intercellular spaces and slower expansion, to a lesser extent. High nitrogen fence ability of cell division, With sufficient nitrogen to meet the post-cell development a long time, Time and maturity length, With the increase of nitrogen, Leaf thickness of the fence, sponge thickness and leaf thickness increased gradually.4. Change of ultra micro structure of different nitrogen of flue-cured tobacco upper leaves during growth and development Time of leaf length 10 ~ 15cm, Tobacco Leaves under Different Nitrogen little difference ultra structure, However, in nitrogen and nitrogen-free high nitrogen vacuolar mature earlier; when the leaves length fixed, more starch granules and osmiophilic globules accumulated with the chloroplast in degradation in high nitrogen, In leaf ripe stage Chlorophyll synthesis is still a constant, Photosynthetic rates relatively stable.
