By taking four types of Phellinus igniarius which came from different areas as research object. The solid medium and culture condition optimization ; liquid medium and culture condition optimization and liquid submerged fermentation culture of Phellinus igniarius was researched deeply separately. The main achievements of our studies are as following:1. Research on solid medium of Phellinus igniarius and the optimization of culture conditionsExperiments were focused on the effect of differernt cell age, carbon sources, nitrogen source, carbon-nitrogen ratios, trace elements and their concentration combinations, and temperatureon on the mycelial growth of Phellinus igniarius.The results showed that: when we sampled in mother culture petri dishes with a radius 3cm and then inoculated, the yield of mycelia was greatest in all of the four kinds of Phellinus igniarius. The most suitable carbon sources, nitrogen source and their concentration combinations were: glucose 3%, peptone4%; trace elements and their concentration combinations were: potassium dihydrogen phosphate0.1%, magnesium sulfate 0.1%,copper sulfate0.2%, vitamin B complex 0.05%, vitamine E 0.05%; the best culture temperature was 25℃.2. Research on liquid medium of Phellinus igniarius and the optimization of culture conditionsAfter investigation the mycelial biomass and polysaccharide accumulation rule of Phellinus igniarius by using liquid culture. Effect of differernt carbon sources, nitrogen source, carbon-nitrogen ratios, trace elements and their concentration combinations, rotation speed, temperature, pH value, medium volume on the yield of exo polysaccharide by using liquid culture was studied in detail respectively.The results showed that: the mycelial biomass and polysaccharide yield of Phellinus igniarius NO.1 and NO.2 reached maximum in the liquid culture for 10and 11 days respectively; while the mycelial biomass and polysaccharide yield of Phellinus igniarius NO.3 and NO.4 reached maximum in the liquid culture for 11 and 12 days respectively.The most suitable formula of liquid culture medium was screened out by taken Phellinus igniarius exo polysaccharide as the final aim in these experiments. Phellinus igniarius NO.1 and NO.2: corn meal 3%, wheat bran2%, potassium dihydrogen phosphate 0.2%, magnesium sulfate 0.05%, Calcium Carbonate0.2%, vitamin B complex 0.05%, vitamine E 0.1%; Phellinus igniarius NO.3 and NO.4: corn meal 2%, wheat bran 2%, potassium dihydrogen phosphate 0.2%, magnesium sulfate 0.05%, Calcium Carbonate0.2%, vitamin B complex 0.05%, vitamine E 0.1%.The results showed that: the most suitable culture condition for the four kinds of Phellinus igniarius strains were basically in accordance. rotation speed was150r/min , temperature was 28℃, PH value was 6.50 , capacity of erlenmeyer flask and medium volume ratio is 250 ml :80ml.3 Research on liquid submerged fermentationculture of Phellinus igniarius After investigation the mycelial biomass and polysaccharide yield accumulation rule of Phellinus igniarius by using liquid submerged fermentation, a stain which has the highest polysaccharose yield was chosen from the four Phellinus igniarius stains. Then the effect of differernt bamboo sawdust addition and dissolved oxygen content in fermentation broth on mycelial biomass, the crude extracellular polysaccharide and extracellular polysaccharide yield.The results showed that: the mycelial biomass and polysaccharide yield of all the Phellinus igniarius strains reached maximum in the fermentation for 3 and 4 days respectively, and the Phellinus igniarius NO.3 has the highest polysaccharide yield. When the bamboo sawdust amount was 0~3%, the mycelial biomass, crude extracellular polysaccharide and extracellular polysaccharide yield of Phellinus igniarius NO.3 increase with the increase of the bamboo sawdust amount , and reached the maximum when the bamboo sawdust amount was 3% , while the bamboo sawdust amount was increased continuously, the mycelial biomass, crude extracellular polysaccharide and extracellular polysaccharide yield decreases with the increase of the bamboo sawdust amount.The mycelial biomass of Phellinus igniarius NO.3 was in direct proportion with the increase of the dissolved oxygen content, but the crude extracellular polysaccharide and extracellular polysaccharide didn’t increase with the increase of the dissolved oxygen content, reached the maximum when the dissolved oxygen content was 30%, then decreases with the increase of the dissolved oxygen content in the fermentation broth.
