In this paper, several sensitive electrochemical DNA biosensors were successfully realized on nanocomposites modified electrodes. The resulted electrochemical DNA biosensors were prepared and used to detect the products of the transgenic plant.The paper is made up of five parts. Firstly, a summary referring to the development of the transgenic plant and the detection methods of the electrochemical DNA biosensor and the application in biosensing of nanocomposites are reviewed. The second part, the third part and the fourth part are the investigation on the role of several multi-walled carbon nanotubes nanocomposites during the course of the immobilization of the DNA probe. The properties of the nanocomposites, the characteristics of the immobilization and hybridization of DNA were studied by cyclic voltammetry, differential pulse voltammetry and electrochemical impedance spectroscopy. The remarkable difference between the Ret value at the DNA-immobilized electrode and that at the hybridized electrode could be used for the label-free EIS detection of the target DNA. The loading of the DNA probe on the nanocomposites films are greatly enhanced and the sensitivity for the target DNA detection is markedly improved based on the synergistic effect origin from the nanocomposites. The sequence-specific DNA of phosphinothricin acetyltransferase (PAT) gene and the polymerase chain reaction (PCR) amplification of nopaline synthase (NOS) gene from transgenically modified beans are determined with this label-free electrochemical impedance spectroscopy. In part five, Au-Pt alloy nanoparticles/polytyramine membranes on the glassy carbon electrode (Au-PtNPs/Pty/GCE) are constructed for the electrochemical sensing of the immobilization and hybridization of DNA. The DNA biosensor has been used to the detection of DNA sequence-specific of phosphoenolpyruvate carboxylase gene (PEP). The conditions for the immobilization and hybridization of the DNA probe are optimized and the biosensors also have good selectivity, stability and reproducibility. At last, the conclusion of the thesis is listed.
