Objective : To Investigate the role of Twist-1 mediated senescence suppression in the process of Cervical Intraepithelial Neoplasia (CIN) developing to cervical cancer, by detecting the expression of Twist-1& CBX-3 in 128 clinical specimens with Immunohistochemistry method.Method:1. One hundred and twenty-eight samples were collected, in which 14 samples with normal cervical tissues, 30 samples with CIN I ~ CIN II, 14 samples with CIN III and 70 samples with cervical cancer. All specimens were divided into 4 groups;2. The expressions of Twist-1 and CBX-3 among groups were detected by immunohistochemistry (SABC) method;3. Specific Twist-1 small interfering RNA (siRNA ) were transfected in SIHA cell of human cervical cancer cell line to down-regulate the expression of Twist-1 and sequentially identify the state of senescence using SA-β-gal staining.Result:1. During the process of normal cervix developing to cervical cancer, the expression of Twist-1 was gradually increased.2. The expression of CBX-3 was increased simultaneously with development of CIN, and it was at the peak in the CIN III, but significantly decreased in the state of cervical cancer formation.3. Being confirmed in vitro test of SIHA cell, the senescence of SIHA cell can be induced by down-regulating the expression level of Twist-1.Conclusion:The Twist-1 mediated senescence suppression plays an antitumorigenic role in inhibiting the cervical cancer formation.
