Abstract

Phytic acid is a natural organic phosphorus compounds, widely present in plant seeds and cereals, legumes and oilseeds. Because of its low toxicity, metal chelation, anti-oxidation significantly, phytic acid is widely used in food, environmental protection, health care, chemical and other industries. Of phytic acid production is the use of acid soluble alkali Shen law, the law steps cumbersome low phytic acid product purity and impurity content is high, there is a big gap and abroad phytic acid products, how to take full advantage of China's rich plant resources for plant acid, and production of the phytic acid products on the market competitiveness of phytic acid industry has a huge significance. Meanwhile, phytic acid in the diet inhibits mineral absorption is usually dose - reactive, low concentrations of phytic acid does not affect the body's absorption of these elements. Therefore, understanding the relationship of of phytate human health, to understand its content in the food intake of the different groups of people, as well as between phytic acid and minerals relationship for guiding the crowd with a reasonable diet, and select the appropriate processing cooking in order to avoid the adverse effects of phytic acid on mineral absorption and utilization of positive significance, at the same time enhance the project went further research, such as food of phytic acid processing and food phytic acid to provide reference. Defatted apple seed as raw material, phytic acid extraction process, the parameters of the anion resin purification phytic acid, antibacterial activity in vitro and in vivo antioxidant activity of phytic acid and phytic acid. (1) phytic acid extraction process test: through the optimization of the selected hydrochloric as phytic acid solvent, select the extraction time, extraction temperature, liquid ratio, acid concentration as a single factor and response surface test, phytic acid extraction rate on The optimal process parameters: temperature 35 ° C, time 0.75h, liquid to solid ratio 17:1 extract solution concentration 0.4mol / L. (2) Test of the anion resin purified phytate showed: LX-300 anionic resin is selected to be purified phytate solution can with 0.7mol/mLNaCl phytic acid completely eluted: elution peak of good symmetry, and the recovery was 90.2 %; low pressure water bath of the collected solution was evaporated to a paste, concentrated phytic acid concentration was measured as 49.45%. (3) phytic acid in vitro anti-oxidation test: phytic acid on DPPH radical · OH radicals and free radical scavenging effect of the O2-· were higher than the VC, the IC50 for 9.31mg/mL, 2.54 mg / ml and 9.28mg/ml. Reducing power of phytic acid than Vc, the inhibition of lipid peroxidation IC50 3.24mg/mL. (4) phytic acid in mice in vivo anti-oxidation tests showed: phytic acid can improve the test serum and liver tissue superoxide dismutase (SOD), catalase (CAT) and glutathione-off peroxidase (GSH-Px) activity, and significantly reduced the tissue content of malondialdehyde (MDA), mouse total antioxidant capacity (T-AOC). (5) antibacterial tests showed: phytic acid and sodium phytate relatively weak inhibitory effect of heat-resistant bacteria Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Bacillus cereus and Bacillus have significant inhibitory effect : For the same species of bacteria, phytic acid inhibitory effect is stronger than the sodium phytate.
