Abstract


Purpose Investigate the mechanism of TGF-?1 induced synthesis of extracellular matrix in skeletal muscle-derived stem cells (MDSC).Method To isolate rat skeletal muscle-derived stem cells by differential adhesion method, medium supplemented with TGF-β1. Cultured cells were divided into three groups, A: control group; B: 10ng/ml TGF-β1; C:10ng/ml TGF-β1+ 10ng/ml CTGF antibody. Cells growth condition was observed under phase contrast microscope. The expression of CTGF, ColⅠand ColⅢwas examined by RT-PCR, Western-blot and immunofluorescence staining.Results In vitro, TGF-β1 can induce MDSC to express CTGF, promote the synthesis and secretion of ColⅠand ColⅢ. CTGF antibody can reduce the expression and synthesis of ColⅠand ColⅢ, partially reverse the effect that TGF-β1 have on the MDSC.Conclusion TGF-β1 can induce the synthesis and secretion of extracellular matrix, thus promoting tissue fibrosis in vitro; TGF-β1-CTGF signaling pathway is one of the mechanisms that mediating TGF-β1 to stengthen the synthesis and secretion of extracellular matrix.
