Objective:To investigate the effect of Livin knockdown on proliferation and migration of cervical cancer cell siha.Methods:After Livin shRNA transfection, viability and migration capability of siha cell were assessed by MTT assays and Transwell migration assay, respectively.Results:Cell proliferation capability and migration capability were inhibited by knocking down Livin (P<0.05).Conclusions:Cell proliferation capability and migration capability of siha cells could be significantly inhibited by RNAi, thus it was a novel target for gene therapy in cervical cancer. Objective:To explore the effect and value of the Livin shRNA in enhancing chemosensitivity of siha cells.Methods: MTT was performed to study sensitivity of the cells to chemotherapy drugs. Cell apoptosis was analyzed by Flow cytometry.Results:After silencing of Livin gene, significant decreases in IC50 were observed in the Livin-knockdown siha cells (P<0.001). Sensitivity of siha cells to chemotherapy drugs (including cisplatin, Taxol and ADM) was markedly increased (P < 0.001).Conclusions:Livin is hopeful to be a molecular target for increasing sensitivity of cervical cancer cell to chemotherapy. Gene silencing mediated by RNA interference may be a new means of gene therapy for cervical cancer.
