Worldwide blood shortage of resources, coupled with emergency surgery, the occurrence of the earthquake, the growing importance of the status of blood substitutes. Hemoglobin blood substitutes as the main raw material, and its activity is closely related to the safety and purity of blood substitutes. The ideal hemoglobin raw material should have a high purity and concentration and is close to the natural activity of the hemoglobin in red blood. The current research results have been reported using different methods, respectively purified hemoglobin solution, but by different degrees of damage to the activity of the purified hemoglobin. Ultrafiltration is a raw active injury smaller means of membrane separation techniques. Based on this characteristic, the experimental introduction of ultrafiltration technology to hemoglobin purification to high purity purified, and close to the natural red blood cells, hemoglobin activity hemoglobin solution. About this study: Objective: To establish and optimize hemoglobin purified ultrafiltration conditions; purified Institute of artificial red blood cells need a high purity and concentration and has natural red blood oxygen carrying capacity of pig hemoglobin solution. And electrophoresis, UV - visible spectrophotometry of purified hemoglobin identified and analyzed by SDS-polyacrylamide gel (SDS-PAGE). In addition, the application of oxygen analyzer hemoglobin solution after ultrafiltration determination and P50 determination of the oxygen-carrying capacity. Results: In the process of ultrafiltration of hemoglobin, the membrane flux with the operating pressure is substantially linear relationship between the increase optimized ultrafiltration conditions were as follows: operating pressure 0.05Mpa, the feed temperature was 12 ° C; Under this condition, the separation and purification of porcine hemoglobin , purity over 99%, the concentration before purification improved about 20 times. Oxygen analyzer, after purification by ultrafiltration pig hemoglobin P50 20.42, close to the natural red blood cell S-type oxygen carrying curve. Conclusion: This topic using ultrafiltration purified porcine hemoglobin, not only to achieve a certain amount of concentration and purity standards, and through the detection of the oxygen-carrying capacity, proved less damage to the activity of the hemoglobin. A further study on this topic ultrafiltration purified hemoglobin our laboratory blood substitutes to provide a better feedstock. Ultrafiltration is a membrane separation technology common, has a non-polluting, under mild conditions, less protein denaturation and recovery high. Therefore, this issue is the method the ultrafiltration purified hemoglobin in the future is expected to be developed industrialized.
