Abstract


To explore Gln-containing tripeptide (Arg-Gly-Gln) can not be weaned piglet intestinal uptake transporters and their weaning piglets Development of this study the 28-day-old weaned piglet jejunum were cultured in vitro, the three tests. The test a 0.56mmol / L Arg-Gly-Gln in weaned piglets isolated jejunum valgus bowel hydrolysis of two treatments (three peptide group and control group). Test Gln / Arg-Gly-Gln, Gly-Gln, free Gln three sources in the uptake transporters in the jejunum valgus gut sac difference made a preliminary study, the incubation time for 40 min, divided into four treatments, namely tripeptide group, the group of the dipeptide, free amino group and the control group (blank Kreb's solution). Test three different concentrations of Arg-Gly-Gln lactate dehydrogenase enzyme activity, intestinal cell proliferation and apoptosis and Gln related metabolic enzyme activity of isolated jejunum culture weaned piglets, divided into six treatments, were added in the culture medium (test group 1) 0 (control group), 0.28, 0.56 (test group 2), 1.11 (test group 3), 2.23 (test group 4) and 4.45 mmol / L (test group 5) Arg-Gly-Gln. The test results are as follows :0-40 min, the Arg-Gly-Gln concentration in the culture medium gradually reduce to a minimum 40 minutes when. The results showed that: 60min incubation time within the Arg-Gly-Gln is not completely hydrolyzed, can exist in complete form. The second test results are as follows: serous fluid and tissue samples, the concentration was significantly higher than that of the tripeptide group of Arg-Gly-Gln and free amino group (P lt; 0.05), group of dipeptide Gly-Gln concentration was significantly higher than The control group and the free amino group (P lt; 0.05); the tripeptide group Gln Absorption Ratio (the mucosal fluid Gln reduced concentration attributable to the ratio of the initial concentration) was significantly higher than the dipeptide group (P lt; 0.05), while the dipeptide group was significantly higher than the free amino group (P lt; 0.05); Gln three forms in the serosal solution of the tripeptide group concentration of higher than that of the dipeptide group (P lt; 0.05), while the group of the dipeptide was higher than free amino group (P lt; 0.05). These results show that: in weaned piglets isolated jejunum Absorption and transport of Gln Arg-Gly-Gln better than Gly-Gln, Gly-Gln better than Gln. The test results are as follows: Compared with the control group, after adding the Arg-Gly-Gln can reduce training lactate dehydrogenase vitality, but no significant change in the protein content of the intestinal tissue; addition to the differences between the treatment group and the control group not significant (P gt; 0.05), while the rest of the treatment group with the Arg-Gly-Gln concentration increases, the increase in the percentage of BrdU-positive cells. Culture correlation coefficient between the concentration of Arg-Gly-Gln, and the percentage of BrdU-positive cells in the liquid was 0.866 (P lt; 0.01). With the increase in the concentration of Arg-Gly-Gln, Caspase-3 the percentage of positive cells was decreased. Correlation coefficient for the culture liquid in a concentration of Arg-Gly-Gln and Caspase-3-positive cell percentage between -0.796 (P lt; 0.01). Control group, treatment group, treatment group 2 between tissue transglutaminase and diamine oxidase activity differences are not significant (P lt; 0.05). Glutaminase activity in the treatment group 3,4,5 intestinal tissue was increased by 27.69%, 46.15%, 83.08%. Treatment groups 4 and 5 intestinal tissue of the treated DAO activity were significantly increased 66.34% and 110.89%, respectively, compared with the control group. In summary: Arg-Gly-Gln in weaned piglets in vitro culture model exists in full form, and can be weaned piglets isolated jejunum uptake transporters in complete form. Moreover, Arg-Gly-Gln may be available through the promotion of weaned piglets from jejunal Glutamine metabolism to promote intestinal cell proliferation, inhibition of apoptosis in, and thus play a role to promote intestinal adaptation.
