Abstract


Purpose: Notch signaling pathway in an evolutionary highly conserved reflect the mechanism of cell communication pathways, which regulate cell proliferation, differentiation and apoptosis, and play an important role in embryonic development and cell fate decisions. The role of Notch signaling pathway in different tissues and cells, even in the same tissue cells in different stages of development role, this physiological role depends on the cell in which the environment. Notch signaling not only for normal cell differentiation plays an important role, played an important role in tumor formation has become a hot issue in recent years, but the mechanism of the formation process of Notch signaling in liver cancer, is not very clear. This study Notch signaling ligands of Notch1 and Jagged1 expression in hepatocellular carcinoma, cirrhotic liver tissue and adjacent normal liver tissues and analysis in HCC clinicopathological parameters, to explore the five proteins the correlation between. Methods: An object of study: 50 cases of hepatocellular carcinoma cancer tissues, corresponding to 50 cases of adjacent cirrhotic liver tissue and 12 cases of hepatic hemangioma surrounding normal liver tissue from the Fourth Hospital of Hebei Medical University, Department of Hepatobiliary Surgery May 2008 to 2009 April surgical specimens. All tissue types were confirmed by the Hebei Medical University Department of Pathology confirmed. All patients were not carried out any form of treatment. 2 methods: tissue removal in 10% neutral formalin-fixed paraffin-embedded spare. Results: 1 Immunohistochemical results showed that Notch1 molecules detected in the liver tissue expression of Notch1 cytoplasm yellow dye. Notch1 expression in HCC associated with sex, age, tumor number, tumor size, AFP, HBsAg, HBeAg negative and positive tumor capsule is complete, whether independent of tumor thrombus; in liver cancer pathological grade well-differentiated cancer tissues expression of Notch1 stronger than poorly differentiated (chi to 2 = 6.938, P = 0.031; r = -0.376, P = 0.007); Notch1 expression and liver cancer clinical stage. 2 of Notch2 were expressed in the liver tissue, the performance of the cytoplasm stained yellow. Notch2 in hepatocellular carcinoma, cirrhotic liver tissue adjacent normal liver tissue, no significant difference in expression between the three. Notch2 expression in HCC associated with sex, age, tumor number, tumor size, AFP, HBsAg, HBeAg negative and positive tumor capsule is complete, whether the tumor thrombus, liver cancer clinical stage and pathological grade were unrelated. Notch3 were expressed in the liver tissue, the performance of the cytoplasm stained yellow. Notch3 expression in HCC was significantly higher than the adjacent cirrhotic liver tissues and normal liver tissue (Z = -2.551, P = 0.011 and Z = -2.436, P = 0.015), cirrhotic liver tissue expression in adjacent normal liver tissue expression No statistically significant difference. Notch3 in HCC was not correlated with gender, age, tumor number, tumor size, AFP, HBsAg, HBeAg negative and positive tumor capsule is complete, whether independent of tumor thrombus; later in liver cancer clinical staging cancer tissue Notch3 was stronger in the earlier stages (chi to 2 = 10.872, P = 0.028; r = 0.359, P = 0.010); Notch3 expression of the degree of liver cancer tumor grade. 4 Notch4 were expressed in the liver tissue, the performance of the cytoplasm stained yellow. Notch4 liver cancer tissues was significantly higher than the adjacent cirrhotic liver tissue and normal liver tissue (Z = -2.906, P = 0.004 and Z = -2.985, P = 0.003), cirrhotic liver tissue expression and adjacent normal liver tissue statistically significant. Notch4 in HCC was not correlated with gender, age, tumor number, tumor size, AFP, HBsAg, HBeAg negative and positive tumor capsule is complete, whether the tumor thrombus, liver cancer clinical stage and pathological grade were unrelated. Jagged1 expression in the liver tissue, the performance of the cytoplasm stained yellow. Jagged1 expression in HCC was significantly higher than the adjacent cirrhotic liver tissues and normal liver tissue (Z = -2.747, P = 0.006 and Z = -2.249, P = 0.025), cirrhotic liver tissue expression and in the adjacent normal liver tissue expression statistically significant. Jagged1 expression in HCC associated with sex, age, tumor number, tumor size, AFP, HBsAg, HBeAg negative and positive tumor capsule is complete, whether the tumor thrombus, are unrelated to liver cancer clinical stage and pathological grade. 6 HCC tissues of Notch1, of Notch2, Notch3, Notch4 and Jagged1 expression correlation: hepatocellular carcinoma expression of Notch1 and Notch2 no correlation (r = -0.003, P = 0.981), with the a Notch3 the expression no correlation (r = 0.166, P = 0.249) with the a Notch4 the expression no correlation (r = 0.184, P = 0.200), with the expression of Jagged1 no correlation (r = -0.248, P = 0.082), the expression of Notch2 and Notch3 no correlation (r = -0.054, P = 0.709), no correlation with the a Notch4 the expression (r = 0.137, P = 0.344) and the expression of Jagged1 no correlation (r = 0.144, P = 0.317), Notch3 and Notch4 expression correlation (r = 0.138, P = 0.339), with the expression of Jagged1 no correlation (r = 0.166, P = 0.249), Notch4 and Jagged1 expression was no correlation (r = 0.245, P = 0.086) Conclusion: 1 of Notch1 Notch3, Notch4 and Jagged1 expression in hepatocellular carcinoma tissue adjacent cirrhotic liver tissue and normal liver tissue increased in adjacent cirrhotic liver tissues and normal liver tissue showed no significant difference. 2 of Notch1 expression in liver cancer pathological grade well-differentiated cancer tissues lower elevated differentiation; Notch3 increased liver cancer later clinical stage cancer tissues compared with earlier installments, suggesting that abnormal activation of the Notch signaling pathway may the occurrence and development of liver cancer.
