Abstract


Objective: intravenous the systemic anesthetic ketamine produce good analgesia without significant respiratory depression, this advantage is suitable in burn patients undergoing anesthesia and field trauma, war and advanced cancer patients analgesic application. Ketamine caused by recent cognitive dysfunction and spiritual side effects adverse reactions, and the elderly are a high risk of postoperative cognitive dysfunction in clinical anesthesia, using ketamine anesthesia may be further increased postoperative cognitive dysfunction in the elderly incidence, thereby limiting the clinical application of ketamine in elderly patients anesthesia. Propofol is commonly used in clinical intravenous anesthetics, many studies have shown that the combination of propofol and ketamine can reduce the amount of ketamine and reduce the excitement of the spirit and cardiovascular side effects, and propofol also has cerebral protective effects, but propofol induced by ketamine impact of recent cognitive impairment is not clear, and no relevant reports. Therefore, the experimental contrast observed the impact of the change of ketamine and propofol and ketamine composite applications after learning of aged rats behavior and hippocampal neurons. Methods: 32 SD aged rats were randomly divided into normal saline group (NS) the ketamine 40mg/kg/h (K) group, propofol 30mg/kg/h (P) group, ketamine 40mg/kg/h isopropyl phenol 30mg/kg/h (PK) group, daily continuous intravenous 2h, administered continuously for seven days. Last 24 hours after administration, the rats in each group by Morris water maze test to find occult station latency and spatial search capabilities. The next day after the water maze test, the rats were sacrificed remove hippocampal tissue Paraffin sections deoxynucleotidyl transfer-mediated nick end labeling (TUNEL) method and immunohistochemical method hippocampal CA1 region neuronal apoptosis and cysteine ??aspartate-specific proteases -3 (Caspase-3) protein expression. Results: ① incubation period (Latency period, LP): with the NS group comparison with the first and second days of LP P group LP no significant change (P gt; 0.05), K and PK group LP was significantly longer (n = 8, P lt; 0.05); compared with the K Group LP, PK group LP shortened significantly (n = 8, P LT; 0.05). LP comparison with the NS group 3, 4, 5 days, P, K, and the the PK set of LP were prolonged (n = 8, P lt; 0.05), compared with the P group LP PK group LP was significantly longer (n = 8, P lt; 0.05), compared with the group K LP, PK group LP shortened significantly (n = 8, P LT; 0.05). ② space contraction capacity: groups through the original platform Views NS group (6.75 ± 1.98) times, P group (4.88 ± 1.13) times, K group (1.88 ± 0.83) times, PK group (3.38 ± 1.19); compared with the NS group, P, K, PK group decreased significantly (n = 8, P lt; 0.05); compared with K, P, PK group increased significantly (n = 8, P lt; 0.05 ); compared with the group P, PK group significantly decreased (n = 8, P LT; 0.05) O. (3) neuron apoptosis: NS group, the apoptosis rate was (1.09 ± 0.21)%, group P apoptosis rate (8.81 ± 1.08)%, group K apoptosis rate (20.42 ± 5.68)%, the PK group apoptosis rate (13.88 ± 1.66)%; compared with NS group, P, K, PK apoptosis rate increased significantly (n = 8, P lt; 0.05); compared with K, P, PK group withered significantly reduced mortality (n = 8, P lt; 0.05); compared with the P group, the PK group apoptosis rate increased significantly (n = 8, P lt; 0.05). ④ Caspase-3 protein expression: NS group (2.41 ± 0.70)%, P group (8.09 ± 1.18)%, K group (17.00 ± 2.11)%, PK group (12.52 ± 1.68)%; NS group comparison, P, K, PK group Caspase-3 expression was significantly increased (n = 8, P lt; 0.05); compared with Group K, P, PK group Caspase-3 expression was significantly reduced (n = 8, P lt; 0.05) ; compared with the P group, PK group Caspase-3 expression was significantly increased (n = 8, P lt; 0.05). Conclusion: ① repeated several times with propofol or ketamine may be addressed the aged rats Recent cognitive dysfunction and hippocampus neuron damage, but the damage of ketamine significantly heavier than propofol. ② propofol ketamine anesthesia can reduce the aged rats cognitive function and hippocampal neuron damage caused.
