Abstract


Objective To investigate the link between the changes of the different stages of diabetic rats Ghrelin and gastric motility abnormalities, to understand the different stages of diabetes Ghrelin gastric emptying. Methods 66 Wistar rats were randomly divided into two weeks diabetic group (DM-2w) and control group (NC-2w), 8-week diabetic group (DM-8w) and control group (NC-8w), 20 weeks diabetic group ( DM-20w) and control disease group (NC-20w). Low dose STZ (STZ, 30 mg / kg) after intraperitoneal injection of insulin resistance induced by high-fat feeding feeding, production of the type 2 diabetic rat model. The first two weeks, eight weeks, 20 weeks after the successful modeling radionuclide method of gastric emptying determination of the stomach half time, 50 minutes stomach radionuclide residual rate; plasma Ghrelin levels by ELISA, immunohistochemistry to observe The gastric tissue Ghrelin immunoreactive cell density. Results 1 DM-2w half time was 22.99 ± 2.53 and 48.76 ± 22.06 NC-2w group compared gastric emptying was significantly accelerated, and the difference was statistically significant (P lt; 0.01). DM-8w half time was 35.24 ± 11.00 minutes, compared with the NC-8w group 43.59 ± 13.79 points, the difference was not statistically significance (P gt; 0.05). DM-20w half-emptying time was 84.02 ± 27.54 minutes, and NC-20w group compared to 47.79 ± 13.22 minutes delayed gastric emptying time, the difference was statistically significant (P lt; 0.01). The difference between the control group without statistical significance (P gt; 0.05) DM-2W rats plasma Ghrelin levels were significantly elevated compared with the NC-2W group difference was statistically significant (P lt; 0.05). DM-8W rats plasma Ghrelin level NC-8W group difference was not statistically significant (P gt; 0.05). DM-20W rats plasma Ghrelin level and NC-20W group and the DM-2W group, the difference was statistically significant (P lt; 0.05). Was no significant difference between the control group (P gt; 0.05). 3 DM-2W rats stomach tissue Ghrelin immunoreactive cells increased with NC-2W group, the difference was statistically significant (P lt; 0.01). DM-8W rats gastric tissue Ghrelin immunoreactive cells NC-8W group difference was not statistically significant (P gt; 0.05). DM-20W rats gastric tissue Ghrelin reduce the number of positive cells with NC-20W group group, the difference was statistically significant (P lt; 0.05). Was no significant difference between the control group (P gt; 0.05). Plasma Ghrelin levels and gastric emptying half-time was a negative linear correlation. The Conclusion 1 diabetes have different forms at different stages of gastric motility changes, in the early acceleration in gastric motility main; With the progression of diabetes, gastric emptying gradually slow down the delay. 2 in the process of change in the gastric motility, the plasma ghrelin also showed a similar trend with the gastric motility changes. Plasma Ghrelin level and gastric emptying half-time was a negative correlation.
