Abstract


Objective: To take advantage of the full gene expression microarray and bioinformatics analysis method to filter the golden hamster buccal mucosa precancerous lesions transformation to squamous cell carcinoma of the important disease genes and pathways . Methods : DMBA induced by the golden hamster buccal mucosa, precancerous lesions and squamous cell carcinoma model , total RNA extraction precancerous lesions and squamous cell carcinoma , the synthesis of single labeled Cy3 fluorescent -labeled cRNA , containing 41,000 genes / ESTs sequences the Agilent rat DNA microarray hybridization Ratio ≥ 2 and Ratio ≤ 0.5 threshold screening differentially expressed genes ; differentially expressed genes row GO (Gene Ontology) functional classification and pathway pathway analysis ; part of the difference by RT-PCR . expressed genes for validation. Results : golden hamster buccal develop from precancerous lesions to the process of the squamous cell carcinoma , were selected to the 1981 differentially expressed genes , including 1037 upregulated genes and 944 downregulated genes , 1861 known genes and 120 unknown genes down 963 up 898 known genes ; GO analysis found that these differentially expressed genes in 14 functional genes involved in metabolism, cell structure ; pathway pathway analysis of nine significant change path in 1861 known differentially expressed genes , 14 genes occur in more than nine pathways on enrichment ; consistent upregulated genes Sparc and down the gene the Casp3 line RT-PCR validation results with the microarray results . Conclusion: The golden hamster buccal mucosa precancerous lesions develop into squamous cell carcinoma is mainly caused by the abnormal expression of Casp3, Cc15, CXCL12, Cc124, Dhfr, Tyms, Vcam1, Vtn, Fn1, Apoe, Alb, Serping1, Sparc and Cyp2b13 gene Caspase- 3 activation pathway , chemotactic factor receptor- adhesion activity pathway , E2F transcription factor regulatory pathways of DNA replication , the G1 / S phase transition related regulatory pathways , cell surface integrin protein interaction pathways , lipoprotein metabolism pathway , platelet cytosolic calcium associated with elevated response pathway of arachidonic acid metabolism pathway and changes in the cytochrome P450 metabolic pathway , eventually leading to the transition from precancerous lesions to squamous cell carcinoma of buccal mucosa . Therefore , targeted for more than 14 gene regulation and for in-depth study of more than nine pathways for treatment and to provide an effective way to prevent oral precancerous lesions to squamous cell transformation .
