Abstract


Objective To investigate the anion exchange protein 2 ( AE2 Anion Exchanger 2 ) differences in expression in normal human villous cells and choriocarcinoma JAR cells , and the channel protein expression changes with choriocarcinoma relationship . ( 1 ) normal human trophoblast cells in primary culture , purification and identification ; human choriocarcinoma JAR cell line culture . Original and incubated normal villus cells is set to the normal group , set the cell lines of human choriocarcinoma JAR choriocarcinoma group . (2) normal trophoblast immunohistochemical method to detect the expression of AE2 on the the JAR cell membrane of cells and choriocarcinoma cell lines . (3) RT-PCR technology to detect the the normal trophoblast the cell and choriocarcinoma JAR cells AE2mRNA expression level . Results (1) In the normal group , normal primary cultured human trophoblast cells can only be observed by the naked eye from the microscope the AE2 's very low expression , resulting photography ineffective . Choriocarcinoma group choriocarcinoma the JAR cell membrane on AE2 -positive cells was ( 91.59 ± 12.10 ) % , much higher than the normal group . (2 ) in the normal villous trophoblast cells and human choriocarcinoma JAR cell lines were detected to AE2mRNA expression . Choriocarcinoma group AE2 OD value of the OD value of the normal group AE2 were ( 0.19 ± 0.01 ) , (0.85 ± 0.21); The the choriocarcinoma group AE2 's OD value significantly higher than the normal group ( P lt; 0.05 ) . The AE2 expression of abnormal raised conclusions , external water and electrolyte balance will be destroyed trophoblastic may be one of the reasons leading to choriocarcinoma occurred .
