P38 Signal Cascades in Hyperosmolarity-induced Mucous Hypersecretion

Abstract


Objective To investigate the p38 mitogen-activated protein kinase (P38) signaling pathways involved in the mechanism of hypertonic stress induces high expression of mucin 5AC (MUC5AC) . Methods 545,695,855,1 055 and 280 mOsm / L hypertonic sodium chloride solution NCl-H292 cultured human airway epithelial cells . 1 280 mOsm / L hypertonic sodium chloride P38 specific inhibitor SB203580 , a specific inhibitor of the c -jun N-terminal kinase (JNK) SP600125 and extracellular signal-regulated protein kinase (ERK) -specific stimulus The inhibitor U0126 intervention factors . By RT-PCR and ELISA observation of cultured cells of MUC5AC transcriptional level and MUC5AC protein levels change . 1 280 mOsm / L hypertonic sodium chloride culture supernatant cell homogenates phosphorylated P38 ( p - P38 ) , phosphorylated JNK (p-JNK) and phosphorylated ERK ( p -ERK ) protein by Western blotting assay level. After the results were 545,695,855,1 055 and 280 mOsm / L hypertonic sodium chloride stimulus , cultured cells the MUC5ACmRNA and protein content was significantly higher ( P lt; 0.01 ) showing osmolality related trends . SB203580 significantly cut 1 280 mOsm / L hypertonic sodium chloride due to the elevated levels of p-P38 , while significantly to cut MUC5ACmRNA and protein content ( P lt; 0.05 ) ; SP600125, significantly reduced the content of p-JNK , and down MUC5ACmRNA transcription and protein content (P lt; 0.05), but the effect is weaker than SB203580 ; 1 280 mOsm / L hypertonic sodium chloride on the content of ERK had no effect ( P > 0.05 ) , corresponding to the U0126 also MUC5ACmRNA transcription and protein content significantly affect (P> 0.05). Conclusion hyperosmotic stress was concentration-dependent manner from the level of transcription induced airway epithelial cells showed mucus hypersecretion , and P38 signaling pathway plays a major role .
