Studies on Pharmacological Effects of Fubao Danggui Jiao of Japan (Ⅱ)

Abstract


Fubao Danggui Jiao (FDJ) of Japan is a kind of Chinese materia medica whose functions are invigorate vital energy and nourishing blood. This paper studied the pharmacological effect on gynecology of two FDJ which were produced with different methods to compare their potency and provide some experimental basis for clinical utilization of the new FDJ.The first part studied the analgesic effect of the two FDJ by two pain models of mice: a chemical stimulation model and a menalgia model. The analgesic effect was evaluated by number of writhing times and latent period. In the chemical stimulation model, the two FDJ had the equivalent analgesic effect, and the different doses didn’t influence their effect. However, in the menalgia model, neither of the two FDJ had the analgesic effect.The second part studied the effect of the two FDJ on some hormone of hypothalami-hypophysis-ovary of rats. Serum FSH, E2, P and LH were measured by ELISA after administration of 14d. FDJ-1 could promote the secretion of FSH with low dose, but inhibit it with high dose. FDJ-1 decreased E2’s concentration with high dose. It can increase the concentration of P and LH. The different doses had the same effect. FDJ-2 promoted the secretion of FSH and LH, inhibited the secretion of P, and did not influence E2.The third part studied the effect of the two FDJ on uterine inflammation of rats. The model was made by a fragment of plastic tube put into the left uterine horns. After administration of 14d, tumescent rates of the uteri were calculated. And the left uterine horns were removed for routine histological examination. The results showed both of the two FDJ had the effect of inhibiting uterine inflammation. Their potencies were equal statistically on the same dose. And FDJ-1 had the same effect with different doses. Histological examination of the left uterine horns indicated that there were dense inflammatory cell infiltration and hyperemia in the blood vessels of the and the normal uterine horns were removed for routine histological examination. The results showed both of the two FDJ had promising therapeutic effects onendometriosis, and their potencies were equal statistically on the same dose. FDJ-1 had the same effect with different doses. FDJ-1 could promote the secretion of FSH, but could not influence that of E2 and P. FDJ-1 could promote the secretion of LH with low dose, but inhibit it with high dose. FDJ-2 increased the concentration of FSH and LH, decreased the concentration of E2, and did not influence P. Histological examination of the ectopic endometria indicated that there were stratified epithelia, and dense inflammatory cell infiltration in the negative control group; while the epithelial cells had atrophied into small cubic cells, and the stroma cells were arranged loosely in the FDJ groups. Histological examination of the normal endometria indicated that there was dense inflammation in the negative control group; while the inflammatory phenomenon was to a lesser degree of the FDJ groups.The last part studied the protective effect of the two FDJ on uteri of mice treated with clomifene. The mice was treated orally with clomifene every morning, and administrated with the two FDJ every afternoon. After 7d, the body weight and the uterine weight were measured, and the uteri were removed for routine histological examination. The results showed the two increased the uterine weight and did not influence the body weight. They could protect the uteri damaged by clomifene. Their potencies were equal statistically on the same dose. And FDJ-1 had the same effect with different doses. Histological examination of the uteri indicated that the endometria were thinnest, the glands were few and small, and the stroma cells were a rranged loosely in the negative control group, while the uteri had no pathological negative control group; while the inflammatory phenomenon was to a lesser degree of the FDJ groups.The forth part studied the effect of the two FDJ on endometriosis of rats. The model was induced by autotransplantation of endometrial tissue onto the abdominal wall. After administration of 30d, serum FSH, E2, P and LH were measured by ELISA. And the volumes of endometriotic implants in each rat were measured. The implants changes in the FDJ groups.
