Abstract


Papers maize seed coat as raw material, using ultrasonic extraction of corn seed coat SDF (soluble dietary fiber) and IDF (insoluble dietary fiber) on the basis of the determination of the chemical composition of its seed coat. For chemical treatment and enzymatic treatment of the IDF is extracted from the corn seed husks, and the use of DEAE Sepharose CL-6B ion-exchange column chromatography, and Sepharose CL-6B gel chromatography, extracted with water as solvent ultrasonic frozen dried The maize seed coat SDF were isolated and purified. Infrared spectroscopy, scanning electron microscopy, X-diffraction, DSC analysis of raw materials and different methods of corn seed coat SDF. The results are as follows: corn seed coat composition determination: moisture content of 6.87%, the starch content of 12.50%, 8.65% protein content, fat content of 0.43%, the IDF content of 32%, the SDF content is 12%. Orthogonal experiment to determine the optimum extraction conditions of ultrasonic extraction IDF and SDF were: particle size of 20 mesh, time 60min, ultrasonic power of 60% and solid-liquid ratio 1:15; particle size of 60 mesh, time 30min, ultrasonic power 70% and solid-liquid ratio 1:35. Orthogonal experiment to determine the chemical treatment and enzymatic treatment IDF to make it into the SDF best treatment conditions were: the soda treatment IDF process as follows: pH 10, temperature 90 ° C, time 2.5h and materials liquid ratio 1:50; the acid treatment IDF best process: pH 5 and a temperature of 100 ° C, time 1.5h and solid-liquid ratio 1:50; the ZSL-B xylanase enzyme treatment IDF best craft : pH 3.0, temperature 50 ° C, the solid-liquid ratio 1:40 and enzyme dosage for 2.0mL / g; the ZSL cellulase treatment IDF the optimum: pH 4.0, enzyme dosage of 1.0 mL / g, time 150min and solid-liquid ratio 1:40; ZSL-1300 cellulases of treatment IDF process: pH 3.0, the enzyme dosage of 1.0 mL / g time 120min and solid-liquid ratio of 1:20. Measured on the physical and chemical properties and antioxidant activity of the different methods of maize seed coat SDF, the results are as follows: the expansion rate of the SDF system of four methods; alcohol extraction drying system SDF water holding capacity; ZSL the the cellulase treatment SDF held the highest oil yield; obtained after ion column chromatography SDF DPPH · radical scavenging effect better; obtained ZSL cellulase treatment SDF better hydroxyl radical scavenging effect ; the SDF obtained after ZSL cellulase better restore. After the purification of water as solvent for the ultrasonic extraction lyophilized prepared SDF by DEAE Sepharose CL-6B was detected at 495nm at a peak; after the sample was obtained after purification through the DEAE Sepharose Cl-6B Sepharose CL-6B purification 495nm detected at the three peaks. Corn on the different methods to testa SDF and the raw material for analysis obtained: sample after the ion exchange column more uniform particles, and preferably accumulation state; water extract lyophilized ion column chromatography was obtained after the SDF preferably crystallinity and heat-sensitive, and the crystallinity of the raw material is relatively weak.
