Abstract


The gingerol and Shogaol , is the main active ingredient in ginger , a large number of studies have shown that both anti-tumor , anti-viral , anti-inflammatory and antioxidant activity of 6 - gingerol and 6 - Shogaol highest activity . Ginger in the separation of the active ingredient is not perfect, the establishment of a fast and efficient method for the separation and purification of great significance . This study, D4020 type macroporous resin, the combination of high-speed countercurrent chromatography and efficient preparative liquid chromatography method , a variety of compounds monomer isolated from ginger . Specific methods: ginger 95% ethanol extract with petroleum ether, defatted, and extracted with ethyl acetate , the ethyl acetate phase D4020 type macroporous resin , washed successively with 25% , 50%, 75% ethanol solution eluted component FrA and FrB. FrA n-hexane - ethyl acetate - methanol - water (10:16:10:10, v / v / v / v) as the solvent system , get the HSCCC-D1200 separated by high-speed countercurrent chromatography the component FrAl and 6 - gingerol ( FRA2 ) . FrA1 then purified by preparative HPLC to give 4 - gingerol . The FrB to n-hexane - ethyl acetate - methanol - water (12:10:15:10, v / v / v / v) as the solvent system , the to get components FrBl after the HSCCC-D1200 separation of high-speed countercurrent chromatography 6 - Shogaol (FrB2) and FrB3. Preparative HPLC purification FrB1 and FrB3 8 - gingerol and 10 - gingerol . In addition , the study also established the flash speed countercurrent chromatography (FHSCCC) Preparation of 6 - gingerol and 6 - the Shogaol step rapid separation of from the ginger ethyl acetate phase . Two-phase solvent system of n - hexane - ethyl acetate - methanol - water (12:10:10:16, v / v / v / v) ratio of the solvent relative to the stationary phase, (12:10 : 10:16 , v / v / v / v ) and ( 12:10:15:10 , with v / v / v / v ) ratio of the lower phase of the solvent as the mobile phase and gradient elution . Using four different types of high-speed countercurrent chromatography (HSCCC-D400, HSCCC-D1200, HSCCC-DL1200 and HSCCC - D9000 ) followed by separation scale amplification, separation the scale mg level zoom to 10g level , up to 95% purity , and the separation time is short .
