Extraction of Polar Active Ingredient from Nature Products with Molecularly Imprinted Polymer

MolecuLarly imprinted polymers (MIPs) can specificallly recognize target compounds and their structural analogues. MIPs can be employed to separate and enrich active ingredients of the natural products, furthermore, they are reusable and economical, which endows MIP have potential application in the separation and extraction of natural products. The purpose of this study is to develop a polar water-soluble MIPs with high selectivity,which as the sepration meduim of preparative chromatography to extract the shikimic acid and quebrachitol from nature products.In this study, we used shikimic acid,α-Methacrylic acid, ethylene glycol methyl acrylate double (EDMA) and azobisisobutyronitrile (AIBN) as the template, monomer, crosslinker and initiator, respectively. Under the condition of the ratio of the above four components being 1:8:8:0.08, we successfuLly obtained the highly selective shikimic acid MIP.The experimental resuLts of the adsorptivity showed that this MIP has strong affinity with its recognition sites:the maximum adsorption apparent capacity was 21.94 nmol/g, and the apparent imprinting effect of K is 5.37. The MIP was also used as the column packing to obtain the MIP-flash column, the capability of the column was evaluated by in-situ low-line chromatography. The resuLts showed that the adsorption capacity is 23.25 mg/g under the conditions of the flow rate being 1.67 mL/min and shikimic acid concentration being 10 mmol/L.14.54% reduction in its adsorption capacity was observed after four times of adsorption-elution recycling, which revealed the excellent reusability of the MIP.We extracted 99.8% purity of shikimic acid from Chinese star anise by the MIP-flash column, which is the foundation of the further industrialization.L-quebrachitol is an important raw material of naturally chiral drugs and is used as a precursor to synthesize many drugs. The covalent complexes quebrachitol MIP was synthesized by the method of inverse suspension polymerization with the glucose as template and 4-phenyl boric acid acrylamide as functional monomer.We have studied the adsorption properties and the mechanism of the molecuLar recognition of MIP.45.34% absorption rate was achieved under the optimum condition of adsorption solvent being pH=11 buffer solution.The strong quebrachitol concentration is relatively high in the waste water from the preparation of rubber. Enormous economic and social effects can be expected to extract white firm veneer from waste water by our method.
