Pear (Pyrus L.) in vitro regeneration system is critical to the success of transgenic pear link. To this end, this study first of all to the three varieties of new early pear pears VII, Zaosu and West Green shoots of the year in early cultured explants from three varieties were screened more appropriate rapid propagation of new varieties pear VII and its profusion of seedlings for the entire study test material, and, at the same time to mature embryos as experimental materials, through to stem, leaf, embryo research, initially established a relatively complete set of pear rapid propagation systems. The main results are as follows: a sterilized explants and primary cultures were collected early pear varieties annual shoots, surface sterilized with 75% alcohol 10s, then 2% sodium hypochlorite solution soak 10 min, sterile water 4 - 5 times. Start using induction medium MS 6-BA3.0 mg / L NAA 0.2 mg / L can be successfully induced stem profusion seedlings. 2 Best Rapid Propagation of screening new varieties of pears VII stem segments in MS 6-BA 3.0 mg / L NAA 0.2 mg / L (treatment 13) medium, clusters of seedlings induced the highest rate. The other two varieties induction rate lower than the new pear VII induction rate. 3 callus induction and proliferation of new pear VII mature embryos and seedlings tufted leaves, stems, etc. as test material callus, which blades (y5) MS 2,4-D 2.0 mg / L 6 -BA 5.0 mg / L NAA 0.4 mg / L callus induction rate was 100%; stems most suitable callus induction medium MS 2,4-D 1.0 mg / L 6-BA 0.5 mg / L, the induction rate 100%; stem segments placed horizontally callus best effect; and mature embryos (p5) 1/2MS 2,4-D2. 0 mg / L 6 - BA 1. 0 mg / L medium, callus induction rate. 4 callus differentiation and proliferation of adventitious buds arising from mature embryo callus treated (13) MS 6-BA 3.0 mg / L NAA 0.2 mg / L culture can induce adventitious buds; proliferation of adventitious buds best the media handle (12) MS 6-BA 1.0 mg / L NAA 0.5 mg / L. 5 explants regenerated plants stems directly induce adventitious buds induced best medium for (12) MS 6-BA1.0 mg / L NAA 0. 5 mg / L, induction rate was 96.34%, multiplication rate was 7.29; different parts of the stem ability adventitious buds from strong to weak upper, middle and lower. 6 rootless seedlings seedling rooting optimum rooting medium for 1/2MS IBA2.5 mg / L sucrose 15g, rooting rate of 90.27%. 7 acclimatization will be rooting plantlets acclimatization time 14 days after sealing transplanted into perlite: soil (1: 1) mixing matrix, transplanting survival rate of up to 85.06%; transplanting time for the July Best .
