Safety Evaluation of Dietary DHA and Three Kinds of Edible Oil on Rats

Abstract


In this paper,the effect of DHApeanut oil,rapeseed oil,soybeen oil on bodyweight,organ coefficientbiochemical indexes and lipid peroxidation in rats wasinvestigated,and the influence of four fatty acids on rats were confirmed,to offerthe data for rational diet of people.SD rats were divided into 9 groups by weightrandomly:control group,low dose of DHA group(300mg/kg),high dose of DHAgroup(1000mg/kg),low dose of peanut oil group(0.225g/kg),high dose of peanutoil group(1.125g/kg),low dose of rapeseed oil group(0.225g/kg),high dose ofrapeseed oil group(1.125g/kg),low dose of soybeen oil group(0.225g/kg),and highdose of soybeen oil group(1.125g/kg).3 monthes later,bodyweight,organcoefficient,biochemical indexes and lipid peroxidation was investigated,part liverfrozen saction was observed under light microscope.Results indicated:body weightof rats with high dose of peanut,rapeseed and soybeen oil were increasedsignificantly compared with control group(P<0.01),bodyweight of rats with lowpeanut oil was also increased(P<0.05).Organ coefficient of liver,enal and testisin high DHA group was higher than control group(P<0.05),organ coefficient oftestis in low peanut oil group was increased(P<0.05),organ coefficient of liver andspleen in high rapeseed oil group was also increased(P<0.05).There was notdifference of heart coefficient in any groups compared with control group.Biochemical indexes indicated:SOD was increased significantly in high dose ofDHA and rapeseed oil group(P<0.05,and DHA group P<0.01),and SOD in lowsoybean group was lower than control group.AST in DHA with low and high dosegroups,and high rapeseed oil group was lower(P<0.05),but ALT in high DHAgroup was increased significantly(P<0.05);TG in high DHA group,low peanut oilgroup and high soybean oil group was decreased significantly(P<0.05);HDL-C inlow dose of rapeseed oil and soybean oil was higher than control group;level ofBUN was higher in low peanut oil group than control group;Cr in high soybean oil group was higher also.Pathological test of liver slices did not have specificchange with HE stain,some slices had a few lipid droplets with oil red stain,itmaybe relevant with containing fat itself,did not indicate fatty liver.There waseffect on bodyweight and organs by over uptaking DHA,peanut oil,rapeseed oiland soybean oil,ALT in high DHA group was increased,indicated liver maybedamaged lightly by high DHA uptakeing,but there was not evident damage orside-effect of all.
