Abstract


[Objective] To establish an in vitro the H.pylori infection PDLFs model to observe H.pylori to acting on PDLFs after its cell proliferation and cell in vitro cultured human periodontal ligament fibroblasts (PDLFs) and Helicobacter pylori (H. pylori) cycle changes to to explore of H.pylori and PDLFs the relationship based on correlation providing laboratory H.pylori and periodontal disease . [ Method ] 1 . Vitro culture PDLFs, the cell concentration was adjusted to 2 × 105/ml . Using a Buchner modified yolk agar selection culture techniques and culture of H.pylori , and adjusting the the H.pylori concentration of 1 × 108cfu/ml , 2 × 107cfu/ml of 4 × 106CFU/ml , 8 x 105cfu/ml 0cfu / ml. 3 each concentration group H.pylori with PDLFs cell co-culture , the establishment of of H.pylori infection PDLFs vitro model using four MTT the (Methyl thiazolyl tetrazolium, MTT) colorimetric assay and flow cytometry ( Flow cytometry, FCM), to observe different concentrations of H.pylori on PDLFs cell growth and proliferation and cell cycle . The [Results] the different concentration H.pylori role PDLFs 6h , 12h, after 24 hours compared with the control group , OD values ??were decreased (p lt; 0.05), different concentration of H.pylori can inhibit PDLFs growth . And acting the same time, with the increase of the concentration of H.pylori OD value gradually decreases; the same concentration , the time is longer , OD value decreases gradually . H.pylori role in PDLFs 12h with H.pylori concentration increases , the proportion of cell cycle GoG1 period gradually increased the proportion of S phase and G2M phase downward trend ( P lt ; 0.05) . [Conclusion ] H.pylori PDLFs proliferation inhibition and cell cycle repression , H.pylori and periodontal disease is closely related .
