Abstract


Objective: To understand the impact of periodontitis on erectile function in rats. Method: 1. 4-week-old healthy male, 10 Sprage-Dawley rats, randomly divided into two periodontitis group (A) and the control group (B), periodontitis group (n = 5), peel the periodontal pocket and of 20 orthodontic wires occlusion of the bilateral maxillary first and second molar teeth neck wire buried in the periodontal pocket, buried 1mg/ml LPS soaked silk establish animal model of periodontitis and periodontal pockets. The control group subjected to the same anesthesia, without any treatment. Each experimental group modeling eight weeks after weighing detect periodontal measured intracavernous pressure (ICP) and the average carotid arterial pressure (MAP). Collected rat blood, maxillary alveolar bone, the corpora cavernosa. The left side of the alveolar bone in turn fixed, decalcification, embedding, sectioning and HE staining, microscopy periodontal inflammation and alveolar bone resorption; removal of the right side of the periodontal soft tissue observed alveolar bone resorption. Serum tumor necrosis factor-α (TNF-α), serum C-reactive protein (CRP) assay kit was measured CRP, TNF-α concentrations, blood determination laboratory to assist in the completion by the Luzhou Medical College Hospital; nitric oxide synthase (NOS) activity kit and cGMP kit determination the corpus cavernosum NOS activity and cGMP concentration; Real-time PCR corpus cavernosum endothelial nitric oxide synthase (eNOS) mRNA expression was measured; Western blot determination of the corpus cavernosum eNOS protein content. 5. Statistical Methods: Data were expressed as mean ± standard deviation (x ± s) SPSS17.0 statistical software to analyze data, periodontitis group and the control group using independent sample t (Independent-Samples T Test) test. Results: 1. Periodontitis group in the clinical and pathological significantly, typical periodontitis performance. Weight and blood sugar A group [322.2 ± 24.88g, 5.38 ± 0.54 mmol / L] than in group B [335.8 ± 19.58g, 5.68 ± 0.94 mmol / L] was no significant difference (P = 0.365, P = 0.552); hemoglobin and red blood cell count than in group B, Group A [160.00 ± 9.92 g / L, 8.71 ± 0.90 x 10E12 / L] [166.00 ± 3.40 g/L.8.97 ± 0.19 x 10E12 / L] no significant difference (P = 0.248, P = 0.564); white blood cell count group A, 13.00 ± 0.93 x 10E12 / L] than in group B [7.01 ± 1.88 × 10E12 / L] was significantly higher (P <0.001) .3.ICPmax/MAP × 100 A group of 3V [9.54 ± 6.16] and 5V [30.91 ± 5.61] compared with group B 3V [30.45 ± 3.12] and 5V [50.52 ± 9.52] were significantly lower (P = 0.018, P lt; 0.001). Serum levels of TNF-α, CRP group A [433.10 ± 69.33pg/ml, 52.67 ± 26.04 ng / ml] than that in group B [208.97 ± 69.23 pg / ml, 22.55 ± 16.36ng/m1] a significant increase (P lt; 0.001 , P = 0.006). 5. Cavernous NOS activity, cGMP concentration A group [1.02 ± 0.48U/mgprot 35.86 ± 11.73 pmol / mg] than in group B [2.95 ± 1.43U/mgprot 50.50 ± 12.17 pmol / mg] significantly significantly (P = 0.037 lower . P = 0.016). 6.eNOS mRNA expression in group A [3.47 ± 1.95] than that in group B [2.54 ± 0.67] no significant difference (P = 0.339). A group of 7.eNOS content [0.58 ± 0.07] compared with group B [0.70 ± 0.08] was significantly lower (P = 0.048). Conclusion: periodontitis can cause eNOS and NOS activity decreased cavernous tissue levels of NO and cGMP concentration decreased, leading to erectile dysfunction in rats.
