Abstract


In this study, using legitimate package : β-CD as the main molecular inclusion Ligustrum oleanolic acid (OLA) , off inclusion and refined separation of its combination of high-speed countercurrent chromatography (HSCCC) , while Ligustrum lucidum water extract vitro antibacterial experiments . Β-CD host molecules , OLA for the preparation of the guest molecules clathrate inclusion complexes were identified by TLC and X-ray at the same time , cooperation with methanol inclusion complex off package . The results show that : the use of electromagnetic stirring inclusion time , inclusion ratio (nβ-CD: n bulk drugs ) 2:1 inclusion temperature 70 ° C , 1h conditions to get the best returns . TLC combined with X-ray identification of inclusion complexes . Thermal extraction using organic solvents to achieve the effect of the better- off inclusion , OLA purity increased from 54.12% to 77.5% . Legitimate use of β-CD package of OLA can play a purification effect . Adduct prepared sample 1 and column chromatography on silica gel sample prepared using HSCCC 2 refined separation , TLC combined the HSCCC screening solvent system . N-hexane - ethyl acetate - ethanol - water ( 16:14:14:5 ) solvent system , photogenic as the stationary phase , lower phase as the mobile phase , HSCCC speed to 900r / m, separation time 400min, share of sample 1 2 OLA purity : 93.8% , 96.4% , respectively . Using adduction combined HSCCC , OLA can play certain refined separation purpose . Antibacterial activities of Ligustrum lucidum water extract used in vitro serial dilution method , determination of the different dilutions of Ligustrum lucidum decoction of Staphylococcus aureus , Escherichia coli and Bacillus Bacillus inhibition . Ligustrum lucidum decoction of Staphylococcus aureus MIC 31 mg / ml , MBC 62 mg / ml ; E. coli MIC of 31 mg / ml , MBC 62 mg / ml ; Bacillus subtilis MIC 4 mg / ml, MBC was 8 mg / ml. Ligustrum having some inhibitory golden staphylococcus aureus , E. coli and Bacillus subtilis .
