Abstract

Objective: To detect blood important redox pairs: a of glutathione (Glutathion) oxidized / reduced form (GSSG / GSH) and coenzyme II (coenzyme II) oxidized / prototype (NADP / NADPH) levels, reflect Western medicine (A group) and the TCM healthy population (B group) the body's oxidation reduction state, and to explore the relationship between the redox state of the Yin and Yang balance. Method: the basic standards of Western medicine and traditional Chinese medicine normal adult health the age 20-29,30-39,40-49,50 over the age of men and women were evaluated; total of 400 people, collected fasting blood 3ml early morning. (1) in the near-neutral conditions dithiothreitol (DTT) the plasma GSSG restore the reduction of GSH, pH8.0. Reduced GSH fluorescence probe o-phthalaldehyde (OPA) shrink synthetic hair strongly fluorescent substance isoindole, improve existing fluorescent spectrophotometric determination of GSH content, more accurate and rapid detection of plasma GSH content. (2) NADPH as the hydrogen delivery body in the phenazine methosulfate (PMS), under the action of methyl-thiazolyl tetrazole (MTT) reduction of A Xu (formazan), and simultaneously generate the NADP again by the glucose-6 - phosphate (G6P) generated under the action of glucose 6 - phosphate dehydrogenase (G6PD) reduction of NADPH MTT reduction Xu A, Xu A productivity with coenzyme II is proportional to the amount of the different stages of this cycle reaction. The use of quantitative spectrophotometric determination of the rate of change of the absorbance values ??of A of Xu on coenzyme II. Results: (1) the same age A, the B group of men and women between the same result: the content of GSH, GSSG, GSH / GSSG ratio, Eh (GSH / GSSG) values, the NADPH content NADP, and NADPH / NADP ratio, Eh (of NADPH / NADP) value was no significant difference (P gt; 0.05). (2) A group and B group of all ages the result is the same: the 20-29 and 30-39 age groups in plasma GSH and GSSG content, GSH / GSSG ratio, Eh (the GSH / GSSG) values, NADPH, NADP content, NADPH / NADP ratio, Eh (NADPH / NADP) value was no significant difference (P gt; 0.05); ages 40-49 and 50 plasma GSH of NADPH was significantly more than the first two ages (P lt; 0.05), GSSG , NADP content, GSH / GSSG ratio, NADPH / NADP ratio, Eh (GSH / GSSG) values, Eh (NADPH / NADP) value than the first two age was significantly higher (P lt; 0.05). And age above 50 than 40-49 ages GSH, lower levels of NADPH, GSSG, NADP content, GSH / GSSG ratio, Eh (GSH / GSSG) values, h (NADPH / NADP) value higher. Conclusions: (1) In this study, the use of fluorescent probes OPA reaction with GSH improved fluorescence spectrophotometric determination of plasma GSH, to make more rapid and accurate determination of the content of GSH in the plasma. (2) the body's yin and yang balance state redox equilibrium closely related to traditional Chinese medicine and Western medicine healthy people of the same age the body maintain a constant redox state, no difference between the male and female gender. (3) 40 years ago, the body's redox equilibrium no significant change after the age of 40 increased with age, antioxidant capacity gradually reduce the redox equilibrium gradually offset to the oxidation direction, suggesting that 40-year-old body begins The turning point of aging.
