Abstract


Carbon disulfide (Carbon disulfide, CS 2 ) as an organic solvent commonly used in industry. In recent years, more and more reports on the influence of the male reproductive system. The preliminary study results show that CS 2 can lead to male rat hypothalamic - pituitary - gonadal (Hypothalamic-of male eel were incubated with TC 199 medium without-gonad axis HPGA) secretory dysfunction also affect testicular tissue nitric oxide (Nitric Oxide , NO) content and total nitric oxide synthase (Nitric Oxide Synthase NOS) activity, inducible NOS (iNOS) activity decreased. The typing NOS expression and expression site in different CS 2 express different exposure groups. 2, NOS activity and NO content: CS 2 treated rats serum total NOS activity in the hypothalamus, pituitary, testicular tissue to reduce NO content decreased; SNP group increased NOS activity, NO content increase; did not change significantly after the injection of L-NMMA. 3, sex hormone levels: CS 2 exposure group serum of GnRH and LH content with the control group declined T content with CS 2 exposure concentration increase there trend first and then decreased FSH content increased with increasing exposure concentration and the high-dose group compared with the control group, the differences were statistically significant (P lt; 0.05); SNP serum of GnRH, LH, and T content of the high-dose group compared to both increased, and the difference was statistically significant (P lt; 0.05), FSH content was not obvious (P gt; 0.05). 4, CS 2 rat sperm quality and NO exposed to the intervention effect: CS reduce 2 exposure group total sperm count, reduced sperm motility, abnormal sperm number (rate) increase, reduce sperm motility. SNP group total sperm count, sperm motility, sperm motility, grade A sperm count has increased. L-NMMA no significant effect on sperm quality. 5, HE staining: CS the hypothalamus neurons karyopyknosis 2 exposure group, part of the neuron cytoplasm vacuolation, edema, and along with the proliferation of glial cells; pituitary eosinophils increased to the high-dose group, the most obvious, some cells appear to nuclear pyknosis, cytoplasmic vacuolization; the spermatogenic tubule atrophy, spermatogenic cell disorder, cell number and level of reduction or even vacuolation, no abnormal stromal changed. SNP group alleviate pathological changes than exposure group; L-NMMA group have aggravated disease trends. Can be seen from the above results: the the immunohistochemistry experiments confirm the CS 2 exposure can affect parts of the amount of each type of NOS expression and expression in the testis tissue; CS 2 exposure can lead to tissue NOS activity and NO content changes, changes in serum levels of sex hormones, changes in the rat sperm quality and the rat hypothalamus, pituitary, testicular tissue pathomorphology; SNP release a certain amount of NO after, to some extent alleviate the CS 2 exposure caused tissue damage and reproductive endocrine disorders, suggesting that CS 2 on the hypothalamic - pituitary - gonadal axis the secretion of reproductive damage is mediated by NO, NO donors to a certain extent, the physical intervention CS 2 toxic effect on the body.
