Objective To investigate the exogenous agrin migration of rat brain cortex astrocytes in vitro mechanical injury . Method 1 . Primary culture of rat cerebral cortex astrocytes , purified and passaged . Copy and astrocytes migration determination of the mechanical scratch : once passaged monolayer cultured astrocytes using pipettes blue turned their guns for \the cell number (Na) and astrocytes distance of scratching the edge of the maximum migration distance ( Dmax ) , to astrocyte migration impact indicators as evaluation agrin . 3 . Observed scratch after 0h, 6h, 12h , 24 h , 48h , 72h time point in vitro cultured astrocytes Na and Dmax value , determining the scratch after astrocytes migrate active period . Different concentrations of exogenous agrin ( 0ng/ml 5ng/ml , 10ng/ml, 20ng/ml , 50ng/ml , 100ng/ml) scratch after 60h observation of damage astrocytes Na and Dmax . On this basis , the selected group of agrin role concentration (5ng/ml, 10ng/ml, 20ng/ml) and after injury at different time points ( 24h , 48h , 60h ) observed agrin migration of astrocytes . The Dmax determination with IPP6.0 software , All data were statistically analyzed using SPSS13.0 software . 48h increased most significantly ( P lt; 0.0001 ) . Scratched 72h astrocytic Na and Dmax values ??than those injured . Scratch after 60h exogenous agrin 5ng/ml 10ng/ml group and 20ng/ml group than in the control group astrocytic Na and Dmax values ??were significantly increased ( P lt; 0.05 ) . Of 100ng/ml group astrocytes Na , and Dmax significantly decreased ( P lt; 0.05 ) . Exogenous agrin 5ng/ml , 10ng/ml, 20ng/ml concentration group and after injury 24h, 48h, 60h, astrocytes Na and Dmax than in the control group was significantly increased ( P lt; 0.05 ) to 10ng/ml concentration group for the most obvious impact of migration of astrocytes ( P lt; 0.05 ) . Conclusion 1 . Astrocytes after mechanical scratching 12h began to migrate significantly increased 48h-72h astrocytoma migration after scratch ; 2 . Exogenous agrin added damage astrocytes can affect migration . Agrin within a certain concentration range (5 ~ 20ng/ml) and duration of action (24 ~ 48h) can significantly promote injury astrocytoma migration ; excessive the agrin role concentration ( eg 50 ~ 100ng/ml) inhibit astrocytes migration.
