Abstract


Objective:The purpose of this study is to compare the effects of low frequence magneticstimulation(MS) deliveried from different level of CNS on expression ofgrowth-associated protein-43 (GAP-43) in spinal neurons of rats after spinal cordinjury(SCI).Methods1. AnimalmodelFifty-one SD female rats were divided randomly into three groups: thetranscranial magnetic stimulation(TMS)group(n=15), the trans-spinalcord magneticstimulationgroupat L1-2(n=15)andthecontrolgroup(n=15),the other 6rats servedas the normal control. Modified Allen’s method was used to produce the spinal cordinjuryattheT9-10level.2. MagneticstimulationMagnetic stimulation were deliveried on rats from the site of crania and L1-2spinal cord respectively since 24h post injury. The dose of MS was 0.5 Hz and 75%ofmaximalintensityoutput(1.9T)ofthemagneticstimulator.Thetrainingprototalincluded 28 sessions of training, with 30 pulses per sessionand one session per day.WhiletheratsinthecontrolgroupwerenotexposedtotheMS.3. OutcomemeasureBBB scale score was choose to measure the motor function of rats. Motionevoked potential (MEP) was used to detecte the integrality of motor pathway.Immunohistochemistryand westernblotanalysis wereusedtodetecttheexpressionofgrowth-associatedproteins-43(GAP-43)protein. 4. StatisticalanalysisThe data was processed by SPSS17.0 .Single factor analysis of variance andTukey ’s post hoc test were used to compare the difference between the two groupswith P<0.05asstatisticalsignificancelevel.Results1. BBBscoreof 4weekspostSPIwas 7.61±0.57incontrolgroup,10.54±0.92in TMSgroupand 12.87±0.46in trans-spinalcordgroup.(P<0.05).2. The amplitude of MEP was obviously decreased and the latent time wasprolongedafterSCI.3. Pathological changes of spinal cord presented the characteristic of chronicincomplete spinalcordinjuryinallgroup exceptinnormalcontrolgroup.4. The GAP-43 expression were mild in the normal spinal cord but increased afterSCI,which were higher in MS groups than that in the control group 4weekspostSCI as detected by immunofluorescence and western blot (P<0.05) ,and washighestinthetrans-spinalcordmagneticstimulationgroup(P<0.05).Conclusion1. Magnetic stimulation could promote nerve regeneration and improve locomotorrecoveryinspinalcordinjuredrats.2. Byusingthelowfrequencemagneticstimulation,thestimulationdelieveriedfromspinal cord gained better locomotor recovery, the mechanisms might be relate totheactivationofcentralpatterngenerator(CPG).
