PCR reaction (Polymerase Chain Reaction, polymerase chain reaction ) is an indispensable means of molecular biology . Conventional PCR techniques have many drawbacks , while using MEMS microfabrication technology to create microfluidic biochip applications in the PCR has a great advantage . Purpose of this study is to design and produce a stable and reliable , low cost, highly integrated functional microfluidic chip , and can work with an external control device combined together to form convenient, fast and efficient PCR amplification system, integration of the DNA fragment sample preparation , amplification and detection . To lay the foundation for further biological applications . The main research contents and results are as follows : 1. Designed a PCR-CE integrated system , which can realize the integration of reagent mixing , PCR reaction , product separation . Microfluidic chip consists of a micro -channel plate and sputtering on the PDMS cover the lower electrode glass substrate bonded . (2) The temperature on the paper designs and sensing electrodes are located under the PCR reaction chamber structure of the finite element thermal simulation analysis . Analysis shows that this design has a better uniform heat distribution , but also has a prominent advantage thermostat . 3 by MEMS technology research , and ultimately determine a set of production PDMS- glass microfluidic chip and reliable technology . SU-8 was prepared using the male die quickly , PDMS of patterns having PDMS microfluidic channel cover plate ; processing the platinum electrode on the glass substrate , and to protect the exposed portion of the electrode , a thin layer of PDMS covering the other parts , to obtain an electrode substrate ; the PDMS cover sheet and bonded to the electrode substrate obtained prepreg has both heating and temperature conductivity electrode and the high voltage electrode CE PDMS- glass chip. 4 built a PCR-CE microfluidic detection systems and procedures related to external devices , and microfluidic systems were tested. Temperature control system heating rate reached 15 ° C / s, fluctuations in the range ± 0.4 ° C. CE system also implements injection , separation and detection .
