Abstract


Objective To detect the GABA-R and Glu-R in different course type Ⅱ diabetic rat model of retinal tissue and positioning explore its occurrence and development of diabetic retinopathy in the mechanism. Methods Wistar female rats in control group and experimental group 15 type Ⅱ diabetic rat model GK rats 15. Grouping for the experimental group-GK group (diabetic group 8W, 16W group, 24W groups of five)-N group and the control group (8W groups, 16W group, 24W groups of five). Adopt Western-blot detection of rat retinal GABA-R and Glu-R protein expression, semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR) detection of rat retinal GABA-R and Glu-R mRNA of immunohistochemistry Detection of rat retinal tissue GABA-R and Glu-R expression positioning. Results 1 Western-blot1) GAB A and Glu compared with controls, diabetic retinopathy, 8W, 16W GABA and Glu increased expression, 24W reduced expression 2) GABA-R compared with the control group, diabetic retinopathy, 8W, 16W GABA-R expression reduce, 24W expression increased 3) Glu-R compared with control group, diabetic retinopathy 8W.24W Glu-R expression decreased, 16W immunohistochemical expression increased 2 1) GABA control group and diabetic retinopathy in rats showed positive expression, the expression sites are mainly located in the retina inner plexiform layer, the core layer of cytoplasm, cytoplasmic outer nuclear layer 2) GABA-R control rats rat retina and diabetic retinopathy showed positive expression, expression sites are mainly located within the network layer of the retina, kernel layer cytoplasmic 3) Glu control rats rat retina and diabetic retinopathy were expressed in expression sites are mainly located in the retinal nerve fiber layer, ganglion cell layer, the core layer of cytoplasm, inner plexiform layer, outer nuclear layer cells in cell pulp 4) Glu-R control rats rat retina and diabetic retinopathy showed positive expression, expression sites are mainly located in the retinal nerve fiber layer, ganglion cell layer, the core layer of cytoplasm, inner plexiform layer; 8W, 24W addition to the above site outside the outer nuclear layer is also expressed in the cytoplasm 3 RT-PCR1) GABA and GABA-R compared with the control group, increased expression of diabetic retinopathy 8W, 16W highest expression, 24W 8W decreased expression of higher than normal group 2) Glu-R Compared with controls, diabetic retinopathy decreased expression 8W, 16W lower expression, 24W was the lowest 3) Glu compared with the control group, the highest expression of diabetic retinopathy 8W, 16W higher expression, 24W lower than the normal group expression of an exciting conclusion neurotransmitters and their receptors GluRl Glu and inhibitory neurotransmitter GABA receptor GABAA-R with the occurrence and development of DR 2 GABA and GABAA-R in the DR early may delay the development of DR; GABAA-R activity Status and DR disease progression 3 GluR1 participate Glu-mediated excitatory neurotoxicity, GluR1 active state with the DR disease progression
