Abstract


Objective: To explore the adjacent tissues in western Henan province the esophageal and its cancer HPV infection and human the chromosome end the enzyme (hTERC gene) expression further clarify the esophageal cancer hTERC gene expression and HPV infection and its clinical significance. Methods: Henan University of Science and Technology from December 2007 to November 2008 fresh frozen specimens of 71 cases of esophageal cancer, turn select the cancerous tissue, cancer tissue and normal tissue cell suspension and wax block. The cell suspension after DNA extraction, PCR amplification, the plasmon resonance to detect esophageal HPV infection, the available test results, including 61 cases of normal tissues, 68 cases of esophageal cancer tissues, 71 cases of cancer tissue. Situation the wax block used in fluorescence in situ hybridization (FISH) to detect hTERC gene expression, the pathological diagnosis hTERC genetic testing including esophageal normal tissues, 45 cases, 27 cases of mild dysplasia organizations, medium / heavy atypical hyperplasia tissue of 22 cases 55 cases of cancer tissue. While the same have hTERC gene and HPV infection test results, including the 48 cases of normal tissues, 39 cases, 42 cases of atypical hyperplasia tissue and cancer tissue. Results of: hTERC gene in normal tissues, Ⅰ grade dysplasia group, Ⅱ / Ⅲ grade dysplasia group, esophageal positive expression rate for 0,25.9%, 54.5%, 90.9% (p lt; 0.05). And hTERC gene Ⅱ / Ⅲ grade dysplasia sensitivity, specificity, positive predictive value, negative predictive value of 54.5%, 90.28%, 63.16%, 86.67%. Dysplasia and esophageal gene copy number was significantly increased, the the atypical hyperplasia average copy number of 2.46, esophageal cancer group was 2.64 of hTERC gene expression is closely related with the level of esophageal lesions. , Esophageal cancer tissue hTERC gene expression and tissue differentiation, depth of invasion, macroscopic type, lymph node metastasis, there was no significant difference (P gt; 0.05). 15 cases of 71 patients with esophageal cancer specimens HPV infection, infection type the 16,18,35,39,51,56,58,66,68,81 high-risk and low-risk types 42, 54; major infection type Do, and the ratio is 18 (6/15), 35 (5/15), 51 (4/15), 42 (3/15). Esophageal cancer in the HPV infection is no obvious correlation with gender, age, height, family history, tobacco and alcohol history, body mass index, and other factors. Normal esophageal tissue, cancer tissue and cancer tissue HPV infection were 4.91%, 11.76% and 7.04%, respectively; pathologically confirmed tissue, normal tissue, atypical hyperplasia tissue, cancer tissue HPV infection were 7.69%, 14.29% and 10.42% were not statistically significant (P gt; 0.05). Adjust the threshold value, the normal tissue and cancer tissue, cancer tissue HPV infection is still not related to sex (P gt; 0.05). Χ ~ 2 test, the cancer tissue joint detection of 48 cases of cancer and 42 cases of atypical hyperplasia tissues were one-sided test P = 0.542, while the two-tailed P = 1.0; atypical hyperplasia tissues χ ~ 2 value 0.025, the esophageal cancer hTERC gene expression and HPV infection do not have sex (P gt; 0.05). Conclusion: The presence of HPV infection in esophageal cancer, mainly in high-risk type. gene with esophageal cancer in hTERC, development is closely related to, and can be used as targets for early diagnosis. And esophageal HPV infection may hTERC gene is still no correlation.
