Association between Va166Met Polymorphism, Serum BDNF Concentration and Severity of Depression in First Episode Major Depressive Disorder

Abstract


Objective To explore the association between BDNF Val66Met polymorphism (Single Nucleotide Polymorphism Database ID:rs6265), serum BDNF concentration and severity of depression in Chinese patients with major depressive disorder (MDD).Methods We included 52 patients with MDD and 69 age-and sex-similar control subjects. The brain-derived neurotrophic factor Val66Met polymorphism and serum BDNF concentration was determined in all participants, and all patients were assessed with the 17-item Hamilton Rating Scale for Depression (HAM-D) for depression severity. We used Pearson correlation or t tests to examine the relationship between the variables if it is compatible. To further evaluate the relationship between serum BDNF levels and the covariates, we conducted covariance analysis to reduce confounding factors.Results Among the MDD(n=52),14(26.9%) with the Val/Val genotype,28 (53.8%) with the Val/Met genotype and 10(19.2%) with the Met/Met genotype were found, while it was 22(31.9%),40(58.0%) and 7(10.1%) in the control subjects, respectively. The Val allele frequencies were 53.8% in patients and 60.9% in health control. No correlations were detected between genotype or allele frequencies and ADD. Mean BDNF serum concentrations from 121 subjects (22.37±5.82ng/ml) were differ between MDD (19.59±6.33 ng/ml) and control subjects (24.46±4.40 ng/ml), Mean BDNF serum concentrations in MDD decreased significantly than control subjects(t=4.753,P<0.01). Mean BDNF serum concentrations among genotypes were differ in total and that were 24.72±4.90,21.57±5.71 and 20.60±6.82ng/ml, respectively. (F=4.673, P<0.05). Stratified by diagnosis, mean BDNF serum concentrations among genotypes were not differ in paitents or control subjects. The concentrations in female were higher significantly than male (23.27±5.76 vs 20.73±5.62, t=2.361, P<0.05). No association was found between severity of depression (HAM-D total score) and BDNF serum concentrations or genotype.Conclusions Val66Met polymorphism was association with local population and ethnic background. And it was no directed association between the genotype and diagnose for MDD. Met carried maybe degrade serum BDNF concentrations which was found in males and was might a risk factor for MDD. No association was found between severity of depression and BDNF serum concentrations or genotype from our inference.
