Abstract

Objective: To study the sperm during capacitation in nitric oxide concentration of dynamic change and its related agents improve sperm quality, effects of nitric oxide in sperm can process, nitric oxide preparations to improve IVF sperm capacitation rate to provide a theoretical basis. Methods: According to the World Health Organization (WHO) criteria for routine semen test results, the semen samples were divided into normal semen group (A) 15 cases of 15 patients with abnormal semen group (B), all semen samples by density gradient centrifugation to obtain sperm Add sperm culture medium was able incubated for 4 h, 37 ° C incubator Sperm nitric oxide detection system able to process nitric oxide concentration changes. In another 10 cases of abnormal sperm group (C), the same approach is divided into two control groups (C 1 ) and the experimental group (C 2 ) average, C 2 30 minutes after the start of detection be nitroprusside, observe the the nitroprusside sperm can process, be baseline basic smooth be NG-methyl-L-arginine, and observe NG-methyl-L-arginine sperm can process parameters and sperm quality. Results: B than in group A at the beginning of detection to the nitric oxide concentration dramatic changes in the average time required for a long time (P lt; 0.05) (T 1 ) appears, and the two groups in the concentration of nitric oxide dramatic changes in nitric oxide concentration (D), no significant differences (P gt; 0.05), duration of the B group than in the group A nitric oxide concentration (T 2 ) significantly increased (P lt ; 0.05), group A and group B T 1 and sperm motility, the linear motion sperm count, sperm motility of linear motion related (P lt; 0.05). A group and B group the dramatic changes of the concentration of nitric oxide, nitric oxide concentration (D) and linear motion related to the sperm count (P lt; 0.05), the A group and B group nitric oxide in the duration of a certain concentration (T 2 ) and sperm motility, sperm viability of linear motion rate, d (very slow or not moving sperm) sperm and sperm whipping frequency ((P lt; 0.05). Conclusions: 1. speculated that sperm capacitation the process is not all sperm capacitation, the vast majority of capacitation in a certain period of time, a small part is distributed in the whole process of capacitation; 2, abnormal sperm group, the concentration of nitric oxide is rapidly changing the time and the emergence of an oxidation nitric oxide after the duration of the high concentration nitrogen drastic changes than normal semen significantly prolonged; 3, the appropriate concentration of nitric oxide can be shortened in the nitric oxide concentration of the abnormal group semen time, NG-methyl-L-arginine can be inhibited semen abnormal group continued state of nitric oxide.
