Abstract


Objective To investigate the intrathecal injection of D- US prop microphone pyridine (Dex) can suppress pain effect of formalin- induced pain in rats , as well as in rat spinal cord dorsal horn of nNOS mRNA expression . This experiment consists of two parts : ( 1) observation of the effects of intrathecal injection of D- dexmedetomidine school rat formalin pain behavior . ( 2) Real Time PCR, to detect the effects of intrathecal injection of D- dexmedetomidine on rat dorsal horn nNOS mRNA expression . 24 adult male SD rats were randomly divided into S group was sham-operated group ( anesthesia , disinfection , not in the soles of the feet injection of 5% formaldehyde , 10 min before anesthesia intrathecal injection of saline 20μl ) ; O group for the surgical control group ; D1 group the the Dex 5μg/kg group ; D2 group the Dex 10μg/kg group . O group , D1 group and D2 group 1.5% isoflurane anesthesia before surgery in all rats right after plantar subcutaneous injection of 5 % formaldehyde 40μl, D1 group and D2 group 10min intrathecal injection of D- US prop microphone piperidine S group and O group intrathecal injection of 0.9 % sodium chloride solution and 20 microliters . Rats in the I phase (O-5min) and phase II (15-50min) pain behavior changes ( including flinching , licking , biting enough action ) , and Dubuisson rated . Rats were sacrificed 1 hour after injection of formalin open laminectomy exposed the lumbar enlargement of the spinal cord , lumbar enlargement of spinal cord was cut with a scalpel blade . Will be collected to the spinal cord tissue is immersed in the non liquid nitrogen preservation solution of the sample RNA was extracted using Trizol method in rat spinal cord tissue total RNA , and protein nucleic acid using a Beckman analyzer measuring the concentration of the total RNA extracted , the extracted total RNA RT into cDNA, 4 ℃ and stored . Real Time fluorescent quantitative PCR method to detect spinal cord dorsal horn the L4-5nNOS mRNA expression . Results of each group of rats , the differences in pain scores in I phase was not statistically significant ( P gt; O.05 ) ; II phase compared with O , S , D1, D2 group rat pain scores significantly reduced , and the difference was statistically significant ( P lt ; O.05 ) , the the D2 group 's pain score of rats was significantly less than the D1 group , the difference was statistically significant ( P lt ; O.05 ) ; O group nNOS mRNA relative expression were significantly higher than the other three groups (P values ??are lt; 0.05 ) , the the nNOS mRNA relative expression of D1 group were significantly higher than the D2 group and S group (P = lt; 0.05 ) . The conclusion intrathecal injection of D- US prop microphone piperidine of formalin - induced pain in rats have significant analgesic effect , with the dose increase and enhance its analgesic effect may be related to inhibition of spinal cord dorsal horn nNOS mRNA expression .
