The Effect of Probiotics and Mesalazine on Ulcerative Colitis in Mice and Their Potential Mechanisms

Abstract


Objective: To observe Lactobacillus acidophilus combined mesalazine in mice with ulcerative colitis (UC) therapeutic effect, and to explore its possible mechanism. Methods: 2.5% DSS 7 天 established animal model of acute UC. The 70 BALB / c mice were randomly divided into seven groups: normal group, model group, negative control (normal saline, NS) group, mesalazine group, Lactobacillus acidophilus low (106CFU/ml), high ( 108CFU/ml) dose group, Lactobacillus acidophilus combined mesalazine groups of 10. Free drinking normal double distilled water, and the remaining six groups of mice free drink 2.5% DSS 7 天 established animal model of acute UC. From the beginning two days before modeling, negative group, mesalazine group, Lactobacillus acidophilus low and high dose group, Lactobacillus acidophilus combined mesalazine group were given sterile saline, mesalazine (30.25mg/ml ), Lactobacillus acidophilus bacteria 1 × 106CFU/ml, 1 × 108CFU/ml, Lactobacillus acidophilus bacteria 1 × 108CFU/ml joint mesalazine (30.25mg/ml) gavage, each 0.2ml, one day two times. Mouse body weight were recorded daily during the experiment, and stool occult blood condition. Modeling the eighth day the mice were killed by cervical dislocation morning off, remove the colon, distal colon tissue taken part formalin-fixed, paraffin-embedded sections, HE staining. Outcomes included: disease activity index (DAI), colon mucosa and histopathological changes visually integration; using immunohistochemical SABC method detects heat shock protein 70 (HSP70) and c-Jun N-terminal kinase (JNK) expression. Results: 1. Drinking DSS, the mice with the number of days increased UC DAI integral constant increase in Lactobacillus acidophilus can reduce DAI points. Lactobacillus acidophilus combined mesalazine group DAI lowest points. Compared with model group, mesalazine group, Lactobacillus acidophilus low and high dose groups were statistically significant (p lt; 0.05). 2 mice in each group UC histological scores were higher than the normal group (p lt; 0.05), mesalazine group, Lactobacillus acidophilus group, Lactobacillus acidophilus combined mesalazine group histological scores decreased, with the model and negative control group, the difference was statistically significant (P lt; 0.05). 3.UC HSP70 expression in mouse colon mucosa was lower than normal (p lt; 0.05), Lactobacillus acidophilus mesalazine treatment, can increase the expression level, the difference was statistically significant (p lt; 0.05). JNK expression in the normal group trace, increased expression of the experimental group, the application of Lactobacillus acidophilus mesalazine treatment, can inhibit its expression, the difference was statistically significant (P lt; 0.05). Conclusions: 1. Lactobacillus acidophilus mesalazine treatment of mice with ulcerative colitis have a role, and both considerable effect; two drug combination works best. 2 acidophilus is effective in treating acute UC mice, which may be related to the inhibition of JNK activation, increased expression of HSP70 in colonic mucosa.
